Abstract: | Eucheuma cottonii merupakan salah satu spesies rumput laut yang banyak dibudidayakan di Indonesia. Rumput laut jenis ini mengandung senyawa fenolik dan flavonoid yang berperan pada berbagai aktivitas hayati, seperti antioksidan. Penelitian ini bertujuan menentukan kadar fenolik dan flavonoid total serta kapasitas antioksidan dengan metode DPPH (2,2-difenil-1-pikrilhidrazil) dan FRAP (ferric reducing antioxidant power) dari ekstrak E. cottonii berdasarkan 3 pelarut pengekstraksi berbeda: air, etanol, dan etil asetat. Dari hasil penelitian didapatkan kadar fenolik total 4,49–53,47 mg ekuivalen asam galat/g ekstrak, kadar flavonoid total 0,16–35,64 mg ekuivalen kuersetin/g ekstrak, kapasitas antioksidan metode DPPH 1,38–6,64 μmol trolox/g ekstrak dan dengan metode FRAP adalah 1,82–39,44 μmol trolox/g ekstrak. Penggunaan pelarut etil asetat untuk mengekstraksi senyawa yang terdapat pada E. cottonii menghasilkan kadar tertinggi pada keempat parameter yang dianalisis. Eucheuma cottonii is a species of seaweed that is widely cultivated in Indonesia. This seaweed contains phenolic and flavonoid compounds that play a role in various biological activities, such as antioxidants. This study aims to determine the total phenolics and flavonoids content and antioxidant capacity of E. cottonii extract using 3 different solvents: water, ethanol, and ethyl acetate. This study used DPPH (2,2-diphenyl-1-picrylhydrazyl) and FRAP (ferric reducing antioxidant power) methods to determine the antioxidant capacity. The results showed that the total phenolic content ranged from 4,49–53,47 mg gallic acid equivalent/g extract, the total flavonoid content ranged from 0,16–35,64 mg quercetin equivalent/g extract, the antioxidant capacity using the DPPH method ranged from 1,38–6,64 μmol trolox/g extract, and using the FRAP method ranged from 1,82–39,44 μmol trolox/g extract. Using ethyl acetate as a solvent for extracting compounds from E. cottonii resulted in the highest content of all four parameters analyzed. |
