| Abstract: | Tumbuhnya kesadaran masyarakat akan manfaat tanaman Tin untuk kesehatan menyebabkan permintaan produk dan bibit tanaman Tin semakin meningkat. Penelitian ini bertujuan mengetahui respon terhadap penggunaan ZPT jenis BAP (6-Benzylamino Purin) dengan IAA (Indole Acetic Acid) terhadap pertumbuhan dan multiplikasi tanaman tin varietas Blue Giant secara in vitro. Penelitian dilakukan di Laboratorium Kultur Jaringan IPB University pada bulan Oktober 2022 hingga bulan Mei 2023. Rancangan percobaan yang digunakan adalah faktorial Rancangan Kelompok Lengkap Teracak (RKLT) dua faktor yaitu BAP (0; 0,5; 1; 1,5) mg L-1 dengan kombinasi IAA (0 ; 0,25; 0,5) mg L-1. Kombinasi BAP 0,5 mg L-1 dengan IAA 0,25 mg L-1 menghasilkan eksplan tertinggi yaitu 3,60 cm pada 8 MST. BAP tunggal yaitu 1,5 mg L-1 menghasilkan jumlah tunas terbanyak yaitu 1,85 tunas. Penggunaan media arang sekam steril dengan teknik penyungkupan pada 4 MST ditahap aklimatisasi menghasilkan rata-rata persentase hidup 91,67%. Public awareness in the health benefits of fig plants has led to an increasing demand for fig products and seeds. This study aims to determine the response to the use of growth regulator type BAP and IAA on the growth and multiplication of Blue Giant fig varieties in vitro. The study was conducted at the IPB University, Tissue Culture Laboratory, from October 2022 to May 2023. The experimental design used was a factorial Randomized Complete Group Design (RCGD) with two factors, namely BAP (6-Benzylamino Purin) (0; 0.5; 1; 1 .5) mg L-1 with a combination of IAA (Indole Acetic Acid) (0 ; 0.25; 0.5) mg L-1. The combination of BAP 0.5 mg L-1 with IAA 0.25 m .L-1 produced the highest explant, namely 3.60 cm at 8 WAP. BAP 1.5 mg L-1, produced the highest number of shoots, namely 1.85 shoots. The use of sterile charcoal media with the cover technique at 4 WAP acclimatization resulted in an average survival rate of 91.67%. |
