| Abstract: | The medaka fish (Oryzias javanicus) is a fish that has the potential to be developed as a model animal to apply the replacement ethics of the 3R concept. This study aims to (a) obtain an optimal protocol for making medaka fish as an animal model of diabetes mellitus (DM) and (b) observe some basic data determining the condition of diabetes mellitus in medaka fish, including growth rate, blood glucose levels, wound healing dorsal fins, behavioral observations, pancreatic histomorphology, and proportion of pancreatic β cells. Induction of diabetes mellitus using streptozotocin with a 35 mg/kg BW dose. Induction was done intraperitoneally five times during three weeks (KAL), once at 19th day (PS1), two times at 12th and 19th days (PS2), and three times at 5th, 12th, and 19th days (PS3). NaCl 0.7% was intraperitoneally applied five times during three weeks (KN). On the 21st day, the caudal fins of all groups were amputated, then wound healing was observed at 24, 48, and 72 h after amputation. The optimal protocol to make medaka fish as an animal model for DM is three times induction using streptozotocin (PS3), which could increase blood glucose levels eight times. The three times induction decreased growth rate, wound healing, changes in swimming movement patterns, the size of Langerhans islet, and the proportion of beta-cells. Ikan medaka (Oryzias javanicus) merupakan ikan yang potensial untuk dikembangkan sebagai hewan model, dalam upaya penerapan etik replacement dari konsep 3R. Penelitian ini bertujuan untuk (a) mendapatkan protokol optimal untuk menjadikan ikan medaka sebagai hewan model diabetes melitus (DM) dan (b) mengamati beberapa data dasar penentu kondisi diabetes melitus pada ikan medaka, antara lain: tingkat pertumbuhan, kadar glukosa darah, penyembuhan luka sirip belakang, pengamatan perilaku, histomorfologi pankreas, dan proporsi sel β pankreas. Induksi diabetes melitus menggunakan streptozotocin dengan dosis 35 mg/kg BB. Induksi dilakukan secara intraperitoneal lima kali selama tiga minggu (KAL), sekali pada hari ke-19 (PS1), dua kali pada hari ke-12 dan ke-19 (PS2), dan tiga kali pada hari ke-5, 12, dan 19 (PS3). NaCl 0,7% diaplikasikan secara intraperitoneal lima kali selama tiga minggu (KN). Pada hari ke-21 sirip ekor semua kelompok diamputasi, kemudian diamati penyembuhan luka pada 24, 48, dan 72 jam setelah diamputasi. Protokol optimal untuk menjadikan ikan medaka sebagai hewan model DM adalah induksi menggunakan streptozotocin (PS3) sebanyak tiga kali yang dapat meningkatkan kadar glukosa darah sebanyak delapan kali, menurunkan tingkat pertumbuhan, penyembuhan luka, pola gerakan berenang, ukuran pulau Langerhans, dan proporsi jumlah sel β pankreas. |
