Abstract: Ikan kembung lelaki (Rastrelliger kanagurta) memiliki nilai ekonomis penting bagi nelayan. Ikan pelagis kecil di WPP-NRI 572 dan WPP-NRI 573 telah mengalami kondisi fully-exploited. Penelitian ini bertujuan memastikan spesies, keragaman genetik dan kekerabatan spesies secara molekuler ikan kembung lelaki (Rastrelliger kanagurta) berdasarkan marka gen 16S rRNA di WPP-NRI 572 dan 573 sebagai dasar pengelolaan perikanan. Penelitian dilakukan pada bulan Juni 2021 hingga Januari 2022. Sampel ikan diambil dari perairan Labuan, Cilacap, dan Banyuwangi dengan jumlah total sebanyak 9 individu. Urutan basa nukleotida hasil sekuensing dianalisis menggunakan software MEGA X. Hasil identifikasi DNA barcoding, menunjukkan spesies ikan kembung dari tiga lokasi berbeda merupakan ikan kembung lelaki (Rastrelliger kanagurta). Nilai keragaman genetik ikan kembung lelaki dari tiga lokasi termasuk dalam kategori tinggi. Hal ini menunjukkan populasi ikan kembung tergolong stok yang baik
The Indian Mackerel has important economic value for fishermen. The small pelagic fisheries in WPP-NRI 572 and WPP-NRI 573 have experienced a fully-exploited condition. This study aims to determine species certainty, genetic diversity and genetic relationships of indian mackerel (Rastrelliger kanagurta) molecularly based on 16S rRNA gene markers in WPP-NRI 572 and 573 as a basis for fisheries management. The research was conducted from June 2021 to January 2022. Fish samples taken from the waters of Labuan, Cilacap, and Banyuwangi with a total of 9 individuals. The nucleotide base sequences of the sequencing results were analyzed using MEGA X software. The identification results of DNA barcoding showed that the mackerel species studied from three different locations were the Indian Mackerel (Rastrelliger kanagurta). The results showed that the genetic diversity value of male mackerel from three locations was included in the high category indicating that the mackerel population was classified as a good stock.
