Sabang Bay and Lhokseumawe waters are part of the Malacca Strait which borders the Indian Ocean which has dynamic oceanographic conditions. Sponges are organisms whose distribution is influenced by oceanographic conditions and water quality. This study aims to examine oceanographic conditions and water quality and their compatibility with the diversity of Demospongiae. Oceanographic parameters and water quality were measured directly in the field at three stations for each location. Demospongiae diversity was identified based on environmental DNA methods. Based on data from Marine Copernicus reanalysis, currents in both waters range from 0.004 – 0.126 m/s and are classified as weak currents. The results of the analysis of the physical-chemical parameters of the waters of Sabang Bay and Lhokseumawe show that both are suitable locations for sponge life. The detected nitrate and phosphate content tend to exceed the optimum range for sponge life. The level of water fertility based on TRIX is classified as oligotrophic to mesotrophic, while based on chlorophyll-a content it is classified as oligotrophic. Based on environmental DNA identification, there are 260 Demospongiae OTUs in Sabang Bay and 148 OTUs in Lhokseumawe. Demospongiae populations in Sabang Bay and Lhokseumawe have moderate diversity with high uniformity and low dominance. Sabang Bay has a better level of suitability for sponge life than Lhokseumawe.id
