Abstract: Aeromonas sp. merupakan bakteri Gram negatif yang bersifat patogenik. Salah satu jenisnya adalah Aeromonas salmonicida dan Aeromonas hydrophila. Kedua bakteri tersebut menyebabkan penyakit furunkulosis dan MAS (Motile Aeromonads Septicemia). Tujuan penelitian adalah menguji patogenisitas bakteri Aeromonas salmonicida dan Aeromonas hydrophila pada ikan lele (Clarias gariepinus). Uji tantang dilakukan pada ikan lele yang memiliki bobot 15,40 ± 1,06 gram dan panjang 9,96 ± 0,61 cm. Uji LD50 dilakukan pada setiap jenis bakteri dengan kepadatan 104 , 105 , 106 , 107 , dan 108 CFU/ekor. Dosis yang digunakan yaitu sebanyak 0,1 mL/ekor. Parameter yang diamati pada penelitian ini adalah LD50, mortalitas, rata-rata waktu kematian, gejala klinis, total eritrosit, total leukosit, hemoglobin dan kualitas air. Uji patogenisitas menggunakan perlakuan kontrol yang diinjeksi PBS, perlakuan injeksi dengan A. salmonicida, perlakuan injeksi dengan A. hydrophila dan perlakuan koinfeksi. Hasil menunjukkan perlakuan ko-infeksi yaitu penginjeksian menggunakan A. salmonicida dan A. hydrophila memiliki tingkat patogenisitas yang lebih tinggi dibanding infeksi tunggal. Mortalitas perlakuan dengan injeksi A. salmonicida dan A. hydrophila sebanyak 66,67% dan rata-rata waktu kematian pada 43,20 jam. Sedangkan, apabila dibandingkan antara bakteri tunggal, bakteri A. hydrophila lebih patogen dibandingkan dengan A. salmonicida dengan nilai pada mortalitas ikan injeksi dengan A. hydrophila sebesar 58,33% dan rata-rata waktu kematian selama 45,26 jam.
Description: Aeromonas sp. is a Gram-negative bacterium that is pathogenic. One type is Aeromonas salmonicida and Aeromonas hydrophila. Both of these bacteria cause furunculosis and MAS (Motile Aeromonads Septicemia). The aim of this study was to examine the pathogenicity of Aeromonas salmonicida and Aeromonas hydrophila bacteria in catfish (Clarias gariepinus). The challenge test was carried out on catfish which had a weight of 15.40 ± 1.06 grams and a length of 9.96 ± 0.61 cm. The LD50 test was carried out on each type of bacteria with densities of 104 , 105 , 106 , 107 and 108 CFU/head. The dose used was 0.1 mL/bird. Parameters observed in this study were LD50, mortality, average time of death, clinical symptoms, total erythrocytes, total leukocytes, hemoglobin and water quality. Pathogenicity test used control treatment which was injected with PBS, injection treatment with A. salmonicida, injection treatment with A. hydrophila and co-infection treatment. The results showed that the co-infection treatment, i.e. injection using A. salmonicida and A. hydrophila had a higher level of pathogenicity than single infection. The treatment mortality by injection of A. salmonicida and A. hydrophila was 66.67% and the average time of death was 43.20 hours. Meanwhile, when compared between single bacteria, A. hydrophila bacteria is more pathogenic than A. salmonicida with a mortality value of fish injected with A. hydrophila of 58.33% and an average death time of 45.26 hours.
