Abstract: Tempuyung (Sonchus arvensis L.) merupakan tumbuhan liar yang termasuk dalam famili Asteraceae. Tempuyung sangat mudah dijumpai di Indonesia dan memiliki potensi sebagai obat herbal. Penelitian ini memiliki urgensi bagi pengembangan ilmu pengetahuan khususnya dibidang fitokimia dan industri farmasi karena potensi obat herbal yang semakin berkembang di Indonesia. Beberapa penelitian sebelumnya telah menginformasikan kandungan metabolit dalam tempuyung, namun belum ada penelitian yang membahas pengaruh proses pengeringan berbeda terhadap kandungan metabolit dan aktivitas antioksidan pada daun tempuyung. Proses pengeringan yang berbeda dapat mempengaruhi komposisi dan konsentrasi senyawa fitokimia dan aktivitas antioksidan daun tempuyung yang berbeda. Sehingga, penelitian ini bertujuan untuk mengetahui profil metabolit dan aktivitas antioksidan daun tempuyung berdasarkan metode pengeringan yang berbeda menggunakan LC-MS/MS dan FTIR. Penelitian ini dilakukan dengan ekstraksi maserasi pada daun tempuyung menggunakan etanol p.a. Rendemen yang diperoleh pada oven, matahari, dan pengeringan udara berturut-turut adalah 11,35 ± 1,47%, 9,01 ± 0,48%, dan 7,08 ± 0,66%. Aktivitas antioksidan ditentukan dengan metode DPPH dan %inhibisi pada konsentrasi ekstrak 100 ppm dengan pengeringan dalam oven, matahari, dan udara masing-masing adalah 48,36%; 45,90%; dan 42,45%. Kemudian dilakukan analisis menggunakan LC-MS/MS dan FTIR untuk mendapatkan data berupa kromatogram dan spektrum metabolit yang terkandung pada masing-masing ekstrak. Terdapat pola yang hampir sama tetapi intensitas yang berbeda menunjukkan adanya perbedaan konsentrasi metabolit pada masing-masing ekstrak. Data tersebut kemudian diolah dengan analisis multivariat principal component analysis (PCA), dengan hasil ekstrak daun tempuyung dikelompokkan berdasarkan metode pengeringan. Pengelompokan ekstrak daun tempuyung berdasarkan perbedaan pengeringan dengan data spektrum FTIR, base peak chromatogram pada LC-MS/MS, dan senyawa terkonfirmasi pada LC-MS/MS menghasilkan plot skor PCA dengan jumlah kedua PC masing-masing sebesar 100%, 52%, dan 86%. Pengembangan database yang digunakan perlu dilakukan guna memperkaya hasil identifikasi senyawa pada daun tempuyung, serta perlunya proses identifikasi metabolit yang berpengaruh terhadap aktivitas biologis daun tempuyung serta korelasinya terhadap metode pengeringan berbeda.
Tempuyung (Sonchus arvensis L.) is a wild plant that belongs to the Asteraceae family. Tempuyung is very easy to find in Indonesia and has potential as herbal medicine. This research has urgency for the development of science, especially in the field of phytochemicals and the pharmaceutical industry because of the growing potential of herbal medicines in Indonesia. Several previous studies have informed the content of metabolites in tempuyung, but there is no research that discusses the effect of different drying processes on the content of metabolites and antioxidant activity in tempuyung leaves. Different drying processes can affect the composition and concentration of phytochemical compounds and antioxidant activity of different tempuyung leaves. Thus, this study aimed to determine the metabolite profile and antioxidant activity of tempuyung leaves based on different drying methods using LC-MS/MS and FTIR. Different drying processes can affected different composition and concentration of phytochemical compounds and the antioxidant activity of tempuyung leaves. This study aims to determine the metabolite profile and antioxidant activity of tempuyung leaves based on different drying methods using LC-MS/MS and FTIR. This research was conducted by maceration extraction on tempuyung leaves using ethanol p.a. The yields obtained on oven, sun, and air drying are 11.35 ± 1.47%, 9.01 ± 0.48%, and 7.08 ± 0.66% respectively. The antioxidant activity was determined using DPPH method and the % inhibition at the extract concentration of 100 ppm by drying in the oven, sun, and air are 48.36%; 45.90%; and 42.45%, respectively. Then an analysis was carried out using LC-MS/MS and FTIR to obtain data in the form of chromatograms and the spectrum of metabolites contained in each extract. There were similar patterns but different intensities indicating differences in the concentration of metabolites in each extract. The data was then processed by multivariate principal component analysis (PCA), with the results of the tempuyung leaves extract grouped based on the drying method. The classification of tempuyung leaf extract based on different drying methods with FTIR, base peak chromatogram on LC-MS/MS, and confirmed compounds on LC-MS/MS resulted in a score plot PCA with the number of PCs being 100%, 52%, and 86%, respectively. The development of the database used needs to be done to enrich the results of the identification of compounds in tempuyung leaves, as well as the need for the identification process of metabolites that affect the biological activity of tempuyung leaves and their correlation to different drying methods.
