Abstract: | Streptococcus pyogenes merupakan bakteri penyebab faringitis dan memiliki mekanisme imunitas antitoksin NAD+ glikohidrolase (IFS), untuk pertahanan diri dari aktivitas NAD+ glikohidrolase (SPN) yang diproduksinya. Dalam penelitian ini IFS dijadikan target reseptor baru sebagai alternatif dalam memecahkan masalah resistensi antibiotik. Pendekatan yang dilakukan dalam penelitian ini adalah menambatkan senyawa-senyawa aktif dari bunga kembang sepatu dan daunnya pada permukaan IFS yang potensial untuk mencegah interaksi antara IFS dan SPN. Penelitian ini menggunakan metode in silico melalui penambatan multi ligan menggunakan program Autodock vina dan Autodock4. Hasil penelitian menunjukkan bahwa tarakserol asetat, stigmasterol, β-sitosterol dan peonidin-3-O- glucoside merupakan senyawa terbaik dengan nilai afinitas -7.5, -7.0, -6.9, dan -6.3 kkal/mol. Keempat senyawa berinteraksi dengan residu Tyr43, Met44, dan His137, dimana ketiganya juga berinteraksi dengan SPN ketika IFS terikat pada ligan alaminya ini. Diharapkan interaksi tersebut dapat mencegah pengikatan IFS terhadap SPN sehingga SPN menjadi toksin aktif yang dapat membunuh sel patogen S. pyogenes. Streptococcus pyogenes is a bacterium that causes pharyngitis and bacterium has an antitoxin NAD+ glycohydrolase (IFS) immunity mechanism, for self-defence from the NAD+ glycohydrolase (SPN) activity it produces. In this study, IFS was used as a new receptor target as an alternative in solving the problem of antibiotic resistance. The approach used in this study was to bonding the active compounds from hibiscus flowers and leaves to surface of potential IFS to prevent interactions between IFS and SPN. This study uses in silico method through computational multi-ligand with the Autodock Vina dan Autodock4 programs. The result showed taraxerol acetate, stigmasterol, sitosterol, and peonidin-3-O-glucoside were the best compounds with an affinity equal to -7.5, -7.0, -6.9 and -6.3 kcal/mol. The four compounds interact with three residues, Tyr43, Met44, and His137, where all three also interact with the SPN when IFS binds to this natural ligand. It is expected that the interaction can prevent the IFS binding of SPN so that SPN becomes an active toxin that can kill pathogenic S. pyogenes. |
