Abstract: Pneumonia adalah salah satu penyakit pernapasan mematikan, khususnya di negara berkembang. Kasus ini semakin marak berkembang beriringan dengan munculnya kasus Covid-19. Penderita Covid-19 dengan pneumonia, diketahui memiliki kadar Interleukin(IL)-6 yang meningkat secara signifikan. Berbagai bahan herbal terbukti mampu menghambat aktivitas IL-6 termasuk daun patikan kebo. Penelitian dilakukan dengan analisis senyawa aktif daun patikan kebo menggunakan instrumen LC-MS dan dilanjutkan dengan metode uji in silico yang meliputi penambatan molekuler dengan aplikasi YASARA, analisis bioavaibilitas dan toksisitas. Hasil analisis LC-MS menunjukkan sebanyak 23 senyawa aktif berhasil diperoleh dari ekstrak etanol daun patikan kebo. Berdasarkan hasil penelitian metode in silico, senyawa neocomplanoside memiliki hasil pengikatan terbaik dibandingkan senyawa aktif daun patikan kebo lainnya terhadap IL-6, yaitu dengan nilai ΔG sebesar -5,386 kkal/mol serta konstanta disosiasi 112,72 μM. Senyawa neocomplanoside juga diprediksi dapat diserap baik oleh tubuh karena telah memenuhi aturan Lipinski dan tidak bersifat toksik, sehingga aman untuk dikonsumsi. Dengan demikian, simpulan penelitian ini senyawa aktif neocomplanoside memiliki kemampuan pengikatan terbaik dengan nilai ΔG lebih besar dari hydroxychloroquine dan L-tartaric-acid, yang merupakan ligan pembanding dan ligan alaminya.
Pneumonia is one of the deadliest respiratory diseases, especially in developing countries. Pneumonia cases are increasingly growing in tandem with the emergence of Covid-19 cases. Covid-19 patients with pneumonia are known to had significantly increased levels of IL-6. Various herbal materials have been shown to be able to inhibit IL-6 activity, including Patikan Kebo leaves. The research was conducted by analyzing the active compounds of Patikan Kebo leaves using the LC-MS instrument, and continued with in silico method, which included molecular docking with the YASARA application, bioavailability, and toxicity analysis. The result of LC-MS analysis showed there are 23 active compounds obtained from ethanol extract of Patikan Kebo leaves. Based on the in silico results, neocomplanoside has the best binding affinity compared to other active compounds of Patikan Kebo leaves that interact with IL-6, with the value of ΔG -5,386 kcal/mol and dissociation constant 112.72 μM. Neocomplanoside is also predicted to be well absorbed by the body because it has complied with the Lipinski test and are not toxic, so it is safe for consumption. Thus, this study concludes neocomplanoside with the best binding ability has a greater ΔG value than hydroxychloroquine and L-tartaric-acid as the comparative and natural ligands.
