Abstract: Nila merupakan salah satu jenis ikan air tawar yang banyak dibudidayakan dan dikonsumsi masyarakat Indonesia karena mempunyai laju pertumbuhan cepat dan menempati posisi strategis di pasar ekspor. Permintaan ikan nila yang tinggi harus diikuti dengan penggunaan pakan yang efisien dalam meningkatkan kualitas reproduksi. Suplementasi analog kurkumin dalam pakan dapat mengoptimalkan kondisi fisiologis dan produktivitas hati ikan nila dalam menyintesis vitelogenin sebagai prekursor kuning telur. Kurkumin bersifat hepatoprotektor pada sel-sel hati selama periode reproduksi sehingga dapat meningkatkan kuantitas produksi vitelogenin sebagai nutrisi untuk perkembangan embrio menjadi larva. Suplementasi analog kurkumin dari minyak lawang ke dalam pakan merupakan suatu cara yang dirancang untuk meningkatkan biosintesis vitelogenin oleh hati sebagai upaya meningkatkan ketersediaan bahan-bahan yang dibutuhkan untuk menopang pertumbuhan dan perkembangan embrio dan larva yang optimum sehingga menghasilkan bibit ikan berkualitas baik. Penelitian dilakukan dalam tiga tahap: 1. Peranan analog kurkumin dalam meningkatkan performa hati untuk mendukung reproduksi ikan nila (Oreochromis niloticus), 2. Perbaikan potensi reproduksi dan kualitas telur ikan nila (Oreochromis niloticus) dengan suplementasi analog kurkumin, 3. Performa pertumbuhan larva dan benih yang dihasilkan oleh induk ikan nila (Oreochromis niloticus) dengan suplementasi analog kurkumin. Penelitian tahap pertama merupakan pemeliharaan induk betina ikan nila merah dengan suplementasi analog kurkumin dalam pakan dan tepung kunyit serta kurkumin murni komersial sebagai pembanding. Pemberian analog kurkumin dalam pakan selama 6 minggu menunjukkan terjadinya peningkatan jaringan hati ikan nila, penurunan konsentrasi malondialdehyde (MDA) hati, serum glutamic pyruvic transaminase (SGPT), dan serum glutamic oxaloacetic transaminase (SGOT) yang secara total menggambarkan peningkatan fungsi hati. Peningkatan fungsi hati melalui suplementasi analog kurkumin akan meningkatkan kapasitas hati untuk menyintesis vitelogenin yang akan disimpan dalam folikel yang sedang berkembang selama kematangan gonad. Penelitian ini menggunakan rancangan acak lengkap dengan tujuh perlakuan dan tiga ulangan. Perlakuan yang diberikan adalah Kontrol (tanpa suplementasi), CA 2,4 (2,4 mg analog kurkumin/100 g pakan), CA 4,8 (4,8 mg analog kurkumin/100 g pakan), TP 25 (25 mg tepung kunyit/100 g pakan), TP 50 (50 mg tepung kunyit/100 g pakan), CC 2,4 (2,4 mg kurkumin murni komersial/100 g pakan), dan CC 4,8 (4,8 mg kurkumin murni komersial/100 g pakan). Penelitian tahap kedua merupakan pemeliharaan induk betina sampai terjadi fertilisasi dan pemijahan telur. Performa pertumbuhan dan reproduksi ikan nila merah yang disuplementasi analog kurkumin menunjukkan peningkatan panjang dan bobot harian, peningkatan fekunditas, nilai gonadosomatik indeks, peningkatan konsentrasi vitelogenin telur, derajat pembuahan, derajat penetasan, dan tingkat kelangsungan hidup. Pemberian analog kurkumin dalam pakan terbukti dapat meningkatkan potensi reproduksi ikan nila merah. Penelitian tahap ketiga merupakan pemeliharaan larva yang dihasilkan dari induk nila merah dengan suplementasi analog kurkumin dalam pakan. Pertumbuhan dan perkembangan awal larva sangat dipengaruhi oleh ketersediaan makanan yang berasal dari kantung telurnya. Pemenuhan nutrisi pada awal masa pertumbuhan dapat memengaruhi perkembangan larva. Pemeliharaan larva tahap ketiga menunjukkan bahwa pemberian kurkumin ke dalam pakan memberikan pengaruh pada panjang badan awal hari ke-0, koefisien keragaman panjang badan, panjang larva kuning telur, panjang dan bobot larva lepas kuning telur, konsentrasi hormon tiroksin, panjang badan larva selama 14 hsp, panjang badan larva selama 28 hari hsp, pertumbuhan panjang dan bobot mutlak, pertambahan panjang dan bobot harian benih, dan konsentrasi hormon testosteron benih ikan nila. Kinerja indukan nila betina selama 45 hari setelah penetasan menghasilkan 1225,42 ekor benih berkualitas baik pada kelompok ikan yang diberi analog kurkumin dosis 2,4 mg/100 g pakan. Dengan demikian suplementasi analog kurkumin dalam pakan dapat melindungi sel hepatosit dari pengaruh stres oksidatif akibat radikal bebas serta meningkatkan performa pertumbuhan dan reproduksi ikan nila merah sehingga menghasilkan larva dan benih berkualitas baik.
Tilapia is one type of freshwater fish that is widely cultivated and consumed by the people of Indonesian because it has a fast growth rate and occupies a strategic position in the export market. The high demand for tilapia should be followed by the use of feed efficiency in improving the quality of reproduction. Curcumin analogue supplementation in feed can optimize the physiological condition and productivity of tilapia liver in synthesizing vitellogenin as a precursor to egg yolk. Curcumin is an agent for hepatoprotection of liver cells during the reproductive period so that it can increase the quantity of vitellogenin synthesis and secretion as a nutrient for embryo and larval development. Supplementation of curcumin analogues synthesized from lawang oil into feed is a way designed to improve the biosynthesis of vitellogenin by the liver in an effort to increase the availability of the ingredients needed to sustain the optimal growth and development of embryos and larvae so that it will produce good quality fish seedlings. This study was conducted in three stages: 1. The role of curcumin analogues in improving liver performance to support the reproduction of tilapia (Oreochromis niloticus), 2. Improvement of reproductive potential and egg quality of tilapia (Oreochromis niloticus) with curcumin analogue supplementation, 3. The growth performance of larvae produced by tilapia (Oreochromis niloticus) with curcumin analogue supplementation. The study used a completely randomized design with seven treatments and three repetitions. The treatments given are Control (without curcumin supplementation), CA 2,4 (supplementation of 2,4 mg analog curcumin/100 g feed), CA 4,8 (supplementation of 4,8 mg analog curcumin/100 g feed), TP 25 (supplementation of 25 mg turmeric powder/100 g feed), TP 50 (supplementation of 50 mg turmeric powder/100 g feed), CC 2,4 (supplementation of 2,4 mg commercial pure curcumin/100 g feed), and CC 4,8 (supplementation of 4,8 mg commercial pure curcumin/100 g feed). The first phase of research is the maintenance of the red tilapia with curcumin analogue supplementation in feed and turmeric powder and commercial curcumin as a comparison. Curcumin analogue supplementation in feed for 6 weeks showed an increase in tilapia liver tissue, decreases in the concentration of liver MDA and SGPT, as well as serum SGOT which in total improved liver function. Improved liver function through curcumin analog supplementation increases the liver's capacity to synthesize vitellogenin to be deposited in the developing follicle during gonad maturity. The second stage of research is the maintenance of the female tilapia until fertilization and spawning of eggs occurs. The growth and reproduction performance of tilapia supplemented with curcumin analogues show an increase in daily length and weight, an increase in fecundity, the gonadosomatic index value, an increase in the concentration of egg vitellogenin, fertilization rate of ovulating oocytes, an increase in the hatching rate of fertilized ova, and the increase in the survival rate of the embryo and larvae. The curcumin analogous supplementation in feed is proven to increase the reproductive performance of red tilapia. The third stage of research is the maintenance of larvae produced from the red tilapia fed ration supplemented with curcumin analogue. The initial growth and development of the larvae is strongly influenced by the availability of food derived from its egg sacs. The fulfillment of nutrients at the beginning of the growing period can affect the development of larvae. The maintenance of the third stage of the larvae shows that the supplementation of curcumin analog into the feed affects and improves the length of the body early in the 0th day, the coefficient of diversity of body length, the length of the yolk larvae, the length and weight of the larvae released from the yolk, the concentration of thyroxine hormone, the length of the larval body for 14 hsp, the length of the larval body for 28 days hsp, the growth of length and absolute weight, the increase in length and daily weight of the seeds, the length of the larvae, and the concentration of testosterone hormone. The performance of red tilapia broods on 45 days after hatching produced 1225.42 good quality fry in the group of fish fed with curcumin analogues at a dose of 2.4 mg/100 g feed. The given treatment of curcumin analogues in feed can protect hepatocyte cells against oxidative stress due to free radicals and improve the growth and reproduction performance of red tilapia fish so as to produce good quality larvae and seeds.
