| Abstract: | Bakteri patogen seperti Listeria monocytogenes dan Salmonella typhimurium dapat mencemari bahan pangan. Penggunaan bahan pengawet alami sebagai bio pengawet merupakan salah satu cara untuk mengurangi pencemaran pangan. Bakteri asam laktat (BAL) merupakan bahan pengawet alami yang banyak digunakan karena dapat menghasilkan beberapa senyawa penghambat pertumbuhan mikroorganisme patogen, seperti asam organik, hidrogen peroksida (H2O2), diasetil, dan bakteriosin. Penelitian ini bertujuan untuk melakukan analisis senyawa yang berpotensi menghambat L. monocytogenes dan S. typhimurium asal bakteri Lactobacillus plantarum IN05 dan L. rhamnosus IN13. Penelitian ini dilakukan dengan beberapa tahap, yakni peremajaan isolat BAL L. plantarum IN05 dan L. rhamnosus IN13, produksi senyawa penghambat pada 18, 36, dan 54 jam inkubasi, pengujian aktivitas kultur sel (KS), supernatan bebas sel (SBS), dan supernatan bebas sel netral (SBSN). Pengukuran kadar asam total, hidrogen peroksida, dan diasetil pada SBS, serta analisis asam organik secara kualitatif dan kuantitatif dengan kromatografi cair kinerja tinggi (KCKT).
Supernatan bebas sel (SBS) memiliki aktivitas penghambatan terbesar dibandingkan SK dan SBSN. Kadar asam tertinggi dihasilkan oleh L. plantarum IN05 dan L. rhamnosus IN13 adalah 22,16 mg/mL dan 18,74 mg/mL pada inkubasi 54 jam. Penurunan pH terjadi seiring dengan lama waktu inkubasi. Kadar hidrogen peroksida tertinggi yang dihasilkan oleh L. plantarum IN05 dan L. rhamnosus IN13 adalah 0,053 mg/mL dan 0,066 mg/mL pada inkubasi 18 jam. Kadar diasetil tertinggi dihasilkan oleh L. plantarum IN05 adalah 0,57 mg/mL pada inkubasi 54 jam. Konsentrasi hambat minimum (KHM) SBS terbaik yaitu pada inkubasi 54 jam jika dibandingkan dengan inkubasi 18 jam dan 36 jam. Konsentrasi hambat minimum (KHM) SBS L. plantarum IN05 dalam menghambat S. typhimurium dan L. monocytogenes adalah 40% dan 50%, sedangkan L. rhamnosus IN13 dalam menghambat kedua bakteri patogen ialah 50%. Asam organik yang dihasilkan oleh L. plantarum IN05 dan L. rhamnosus IN13 berdasarkan hasil pemisahan HPLC diketahui mengandung asam tartarat, asam laktat, asam asetat, dan asam sitrat yang dibuktikan dengan munculnya puncak asam organik pada waktu retensi sekitar 1,4-3,7 menit. Asam laktat dan asam asetat memiliki puncak yang diketahui dengan waktu retensi pada 2,289 menit dan 2,503 menit dengan nilai konsentrasi 21423,98 mg/L dan 2812,13 mg/L dihasilkan oleh L. plantarum dan 2,326 menit dan 2,485 menit dengan nilai konsentrasi 12007,79 mg/L dan 3502,42 mg/L dihasilkan oleh L. rhamnosus. Senyawa penghambat bakteri patogen dari supernatan bebas sel (SBS) yang dihasilkan oleh Lactobacillus plantarum IN05 dan L. rhamnosus IN13 berpotensi menghambat Listeria monocytogenes dan Salmonella typhimurium. Pathogenic bacteria such as Listeria monocytogenes and Salmonella typhimurium were contaminating food. The use of natural preservatives as bio preservatives is one way to reduce food contamination. Lactic acid bacteria (LAB) are a biopreservative agent that is widely used because they can produce several compounds that inhibit the growth of pathogenic microorganisms, such as organic acids, hydrogen peroxide (H2O2), diasetil, and bacteriocins. This study aimed to analysis compounds that have potential as inhibitors of L. monocytogenes and S. typhimurium from Lactobacillus plantarum IN05 and L. rhamnosus IN13. This research was carried out several stages, starting from re-culturing the LAB bacterial isolates L. plantarum IN05 dan L. rhamnosus IN13, production of inhibitory compounds at 18, 36 and 54 hours of incubation, activity of cell culture (CC), cell free supernatant (CFS) and neutral cel free supernatant (nCFS) was measured, and to determine of organic acids in the CFS qualitatively and quantitatively by high performance liquid chromatography (HPLC). Cell-free supernatant (CFS) had the greatest inhibitory activity compared to CC and nCFS. The highest acid levels produce by L. plantarum IN05 and L. rhamnosus IN13 IN13 were 22.16 mg/mL and 18.74 mg/mL at 54 hours incubation. The decrease in pH occurred along with the incubation time. . The highest levels of hydrogen peroxide produced by L. plantarum IN05 and L. rhamnosus IN13 were 0.053 mg/mL and 0.066 mg/mL at 18 hours incubation. The highest diasetil level produced by L. plantarum IN05 was 0.57 mg/mL at 54 hours incubation. The best minimum inhibitory concentration of CFS was incubated for 54 hours when compared to 18 hours and 36 hours of incubation. The minimum concentration of CFS L. plantarum IN05 in inhibiting S. typhimurium and L. monocytogenes were 40% and 50%. Meanwhile, the concentrations of CFS L. rhamnosus IN13 in inhibiting both pathogenic bacteria was 50%. Organic acid produced by L. plantarum IN05 and L. rhamnosus IN13 based on the results of HPLC separation were known that contain tartaric acid, lactic acid, acetic acid and citric acid as evidenced by the appearance of organic acid peaks at retention of about 1.4-3.7 minutes. The lactic acid and acetic acid peaks are known to appear at retention times were 2.28 minutes and 2.50 minutes with a concentration value of 21423.98 mg/L and 2812.13 mg/L in L. plantarum IN05 and 2.326 minutes and 2.485 minutes with a concentration value of 12007.79 mg/L and 3502.2 mg/L in L. rhamnosus IN13. The compounds inhibiting pathogenic bacteria from the cell-free supernatant (SBS) produced by L. plantarum IN05 and L. rhamnosus IN13 have the potential to inhibit Listeria monocytogenes and Salmonella typhimurium. |
