Abstract: Budidaya ikan lele tanpa pergantian air mengakibatkan amonia terakumulasi dalam media budidaya. Media pemeliharaan dengan kandungan amonia yang tinggi dapat bersifat toksik bagi ikan dan menyebabkan kadar amonia darah ikan menjadi tinggi yang mengakibatkan stres oksidatif sehingga akan mempengaruhi penurunan konsumsi pakan, penurunan kinerja sistem pencernaan, kinerja hati bahkan penurunan performa pertumbuhan ikan. Upaya peningkatan performa pertumbuhan ikan pada kondisi ini dapat dilakukan melalui suplementasi bahan tambahan alami seperti temulawak. Temulawak diketahui mengandung kurkuminoid, xanthorhhizol, turmerone yang berfungsi sebagai antioksidan, antibakteri, kolagoga dan hepatoprotektor. Penelitian ini bertujuan untuk mengevaluasi pengaruh suplementasi temulawak (Curcuma xanthorrhiza) terhadap kinerja pertumbuhan dan status antioksidan benih ikan lele (Clarias gariepinus ) pada sistem budidaya tanpa pergantian air. Penelitian ini menggunakan rancangan acak lengkap (RAL) yang terdiri atas empat perlakuan dengan empat ulangan. Perlakuan yang digunakan yaitu pakan tanpa suplementasi temulawak sebagai kontrol dan tiga jenis pakan dengan suplementasi tepung temulawak yang berbeda yaitu T0,5 (0,5% kg-1 pakan ), T1,0 (1,0% kg-1 pakan ), T1,5 (1,5% kg-1 pakan). Benih ikan lele dengan ukuran 1,69±0,10 g sebanyak 100 ekor ditebar secara acak pada 16 unit bak fiber dengan ukuran 1×1×1 m dengan volume air sebanyak 600 L. Pemeliharaan ikan dilakukan tanpa pergantian air selama 70 hari dan diberi pakan perlakuan secara at satiation dengan frekuensi pemberian pakan sebanyak dua kali sehari pada pukul 08.00 WIB dan 17.00 WIB. Kondisi kualitas air selama pemeliharaan antara lain suhu 25,1-28,3°C. pengukuran suhu dilakukan setiap hari selama pemeliharaan pada pagi dan sore hari menggunakan termometer, sedangkan nilai pH air selama pemeliharaan yaitu 6,6- 7,9. Pengukuran nilai pH dilakukan setiap hari pada pagi dan sore hari menggunakan pH meter digital. Pengukuran nilai oksigen terlarut di lakukan sebanyak 1 kali dalam seminggu selama pemeliharaan, adapun nilai oksigen terlarut selama pemeliharaan yaitu 0,7-1,34 mg L-1. Pengukuran nilai total amonia nitrogen (TAN) dilakukan sebanyak 1 kali dalam seminggu selama pemeliharaan, sampel air kemudian diuji dan dihitung sehingga nilai TAN selama pemeliharaan yaitu 4,27- 64,1 mg L-1. Parameter uji yang diamati meliputi kinerja pertumbuhan (bobot akhir, jumlah konsumsi pakan, rasio konversi pakan, laju pertumbuhan harian, retensi protein, retensi lemak, dan tingkat kelangsungan hidup), aktivitas enzim pencernaan (aktivitas enzim protease, lipase dan amilase), kinerja hati (kondisi jaringan hati, superoxide dismutase dan nilai hepatosomatic index) serta kimia darah (high density lipoprotein, low density lipoprotein, total kolesterol darah dan trigliserida darah). Hasil penelitian menunjukkan adanya peningkatan performa pertumbuhan ikan lele setelah diberi pakan dengan suplementasi temulawak terhadap nilai FCR,laju pertumbuhan harian, nilai retensi lemak dan tingkat kelangsungan hidup ikan pada perlakuan T0,5. Aktivitas enzim pencernaan diantaranya aktivitas enzim amilase pada perlakuan suplementasi temulawak terlihat lebih rendah dari pada kontrol (p<0,05). Sementara aktivitas enzim lipase pada perlakuan T0,5 dan T1,0 lebih tinggi daripada kontrol (p<0,05), namun enzim protease menunjukkan hasil yang tidak berbeda nyata. Berdasarkan analisis polonomial ortogonal, penambahan temulawak pada pakan dengan dosis berbeda menghasilkan nilai LPH dengan pola kuadratik dengan persamaan y = -0,3172x2 + 0,4066x + 1,2445 Dari persamaan tersebut, diperoleh bahwa dosis suplementasi temulawak untuk menghasilkan LPH maksimal yaitu 0,39 %, sehingga diduga nilai LPH optimal yaitu 0,39%. Aktivitas enzim SOD tertinggi terdapat pada ikan dengan perlakuan T0,5 (p<0,05) serta kondisi jaringan dan persentase hati merah dan pucat pada perlakuan T0,menunjukkan hasil yang berbeda nyata dibanding kontrol. Hal ini menunjukkan kondisi jaringan hati yang lebih baik dibanding kontrol. Transportasi nutrien yang optimal setelah pakan disuplementasi juga ditinjau berdasarkan kondisi kimia darah ikan. Total kolesterol, trigliserida darah dan LDL pada pakan perlakuan memiliki nilai yang lebih rendah dibanding kontrol, sedangkan HDL pakan perlakuan T0,5 dan T1,0 menunjukkan nilai yang lebih tinggi dan berbeda nyata dibanding kontrol. Setiap parameter kimia darah, perlakuan T0,5 menunjukkan hasil yang terbaik dibanding perlakuan lainnya. Penelitian ini menyimpulkan bahwa suplementasi temulawak pada pakan dengan dosis 0,4% mampu meningkatkan performa pertumbuhan ikan lele.
Catfish culture with zero water exchange causes an accumulated ammonia in the maintenance media. High ammonia level in the maintenance media is toxic for fish, which causes high ammonia level in fish blood and oxidative stress, thereby decreasing the feed intake, digestive system performance, liver performance, and growth. An effort to improve the fish growth performance due to this condition can be obtained through dietary natural additive material supplementation such as Javanese tumeric Javanese turmeric has known to contain curcuminoid, xanthorhhizol, and turmeron, functioned as antioxidant, antibacteria, cholagogue, and hepatoprotector. This study aimed to evaluate the effect of Javanese turmeric (Curcuma xanthorrhiza) dietary supplementation on growth performance and antioxidative status of African catfish (Clarias gariepinus) in a zero-water exchange culture system. This study used a completely randomized design containing four treatments and four replications. The treatments were non-supplemented diet as a control treatment and three different Javenese turmeric powder supplemented diets at 0,5% kg-1 diet (T0,5), 1,0% kg-1 diet (T1,0), and 1,5% kg-1 diet (T1,5). The 100 catfish fingerlings at 1,69±0,10 g average weight were stocked in 16 fiber tanks at 1 × 1 × 1 m size and 600 L water volume. Fish were maintained with zero water exchange and fed with the treatment diets for 70 days in apparent satiation twice a day at 08.00 and 17.00 (GMT +7). The water quality during maintenance period was remained at 25,1-28,3°C for temperature. Temperature was measured every day during maintenance period in the morning and afternoon with a thermometer. The pH level was 6,6-7,9, which was measured every day in the morning and afternoon with a digital pH meter. The dissolved oxygen level was measured once a week during maintenance period at 0,7-1,34 mg L-1. The total ammonia-nitrogen (TAN) was measured once a week during maintenance period by taking the water maintenance media samples for further test, which then obtained the TAN value at 4,27-64,1 mg L-1. The parameters observed contained growth performance (final weight, diet intake, FCR, specific growth rate, protein retention, lipid retention, and survival rate), digestive enzyme activity, liver performance (liver histology, superoxide dismutase enzyme level, and hepatosomatic index), and blood chemical (high density lipoprotein/HDL, low density lipoprotein/LDL, total cholesterols, total triglycerides). The results showed a significant increase on growth performance of catfish after feeding with the supplemented diets based on the final weight, specific growth rate, and survival rate parameters in the T0,5 treatment. For digestive enzyme activity, amylase enzyme obtained a lower level in the supplemented diet treatments than the control diet treatment (p<0,05). Meanwhile, the lipase enzyme in the T0,5 and T1,0 treatments was higher than the control treatment (p<0,05). Nevertheless, the protease enzyme obtained no significant differences among the diet treatments. Based on the polynomial orthogonal analysis, different dietary supplementationdoses of Javanese turmeric produced an SGR quadratic pattern value with the equation: y = = -0,3172x2 + 0,4066x + 1,2445. From the following equation, the dietary Javanese turmeric supplementation dose for maximum SGR value was 0,39%. Therefore, optimal SGR value is suspected at 0,39%. The highest SOD enzyme activity level in was obtained from the T0,5 treatment (p<0,05), followed by an improved liver histological condition and a higher pale-normal ratio percentage than the control treatment. This condition indicates that the fish liver histological condition in the supplemented diet treatments is better than the control diet treatment. The optimum nutrient transportation in fish after feeding with the diet treatments were also investigated based on the fish blood chemical conditions. The total cholesterol and LDL levels obtained lower values in the supplemented diet treatments than the control diet treatment, while the HDL level in the T0,5 and T1,0 treatments was higher than in the control treatment. Based on all blood chemical parameters, the T0,5 treatment obtained the best parameter results compared to other treatments. This study concludes that the Javanese turmeric supplemented diet at 0,4% dose can improve the growth performance of African catfish.
