Opudi fish (Telmatherina sarasinorum) is an endemic fish species inhabiting Lake Matano, especially in littoral areas. Almost a decade no update regarding the reproduction biology aspects of this fish, may there is a change in their reproduction biology aspects. The aim of the study was to analyze the reproduction aspects of opudi fish Telmatherina sarasinorum in Lake Matano, South Sulawesi. Sampling method was carried out using simple random sampling method. The study was conducted in September 2020-May 2021, of which 1760 individual fishes from 2122 individual fishes, consisting of 1212 males and 548 females were obtained. The results showed that sex ratio of opudi fish in nature was unbalanced. Size of the first gonad maturity of female fishes is smaller than male fishes. Reproductive potential shown by eggs number ranged between 80-507 eggs, with 228 eggs in average in batch, this species is categorized into partial spawning. Spawning ground is presumed in S. Lawa, P. Salonsa, Utuno, and Tanah Merah or around the southern region of Lake Matano. The fish spawning all the time, with peak spawning season in April, July, September and December.
