Title: Profil Leukosit Tikus Hiperkolesterolemia yang Disuplementasi Polisakarida teripang H. nobilis.

Abstract: Hiperkolesterolemia meningkatkan enzim xanthonine oxidoreduktase (XO) penghasil radikal bebas yang berperan timbulnya inflamasi, atherosklerosis, penuaan, iskemia, hemolisis, stroke hingga kematian. Penelitian ini bertujuan menganalisis pengaruh polisakarida H. nobilis sebagai antihiperkolesterolemia terhadap profil leukosit tikus hiperkolesterolemia. Penelitian ini dirancang dengan membagi 12 ekor tikus jantan strain Sprague Dawley menjadi dua kelompok. Kelompok pencegahan terdiri dari kelompok K+ yaitu tikus diberi pakan kolesterol 1% selama 28 hari dan Kelompok PCh yaitu tikus diberi pakan kolesterol 1% dan polisakarida H. nobilis dosis 400 mg/kg BB secara bersamaan selama 28 hari. Kelompok pengobatan terdiri dari kelompok Ch yaitu tikus diberi pakan kolesterol 1% selama 28 hari pertama dan dilanjutkan dengan pakan standard pada 28 hari berikutnya dan kelompok ChP yaitu tikus diberi pakan kolesterol 1% selama 28 hari pertama dan dilanjutkan dengan pemberian polisakarida H. nobilis 400 mg/kg BB selama 28 hari berikutnya. Parameter hematologi yang dianalisis adalah leukosit, limfosit, neutrofil, monosit, eosinofil, dan basofil. Analisis data menggunakan Analisis one way ANOVA dilanjutkan uji Duncan. Hasil penelitian menunjukkan bahwa jumlah leukosit pada kelompok K+ adalah sebanyak 8883±2413 butir/mm3, Pch sebanyak 7883±728 butir/mm3, Ch sebanyak 5900±1829 butir/mm3, dan ChP sebanyak 3950±975 butir/mm3. Jumlah limfosit pada kelompok K+ adalah sebanyak 6603±892 butir/mm3, PCh sebanyak 5070±2479 butir/mm3, Ch sebanyak 4130±1776 butir/mm3, dan ChP sebanyak 2435±982 butir/mm3. Jumlah neutrofil pada kelompok K+ adalah sebanyak 2601±2258 butir/mm3, PCh sebanyak 2161±994 butir/mm3, Ch sebanyak 1593±1776 butir/mm3, dan ChP sebanyak 1448±927 butir/mm3. Jumlah eosinofil pada kelompok K+ adalah sebanyak 0±0 butir/mm3, PCh sebanyak 105±451 butir/mm3, Ch sebanyak 59±146 butir/mm3, dan ChP sebanyak 26±79 butir/mm3. Jumlah basofil pada kelompok K+ , PCh, Ch, dan ChP adalah sebanyak 0±0 butir/mm3. Pemberian polisakarida teripang H. nobilis berpengaruh nyata terhadap jumlah leukosit, Kelompok yang diberi polisakarida teripang H. nobilis jumlah leukosit lebih rendah daripada kelompok yang tidak diberikan polisakarida teripang H. nobilis. Polisakarida teripang H.nobilis lebih efektif digunakan sebagai pengobatan dari pada pencegahan. Pemberian polisakarida teripang H. nobilis berpengaruh nyata terhadap jumlah jenis-jenis leukosit.
Hypercholesterolemia increases the activity of the enzyme xanthonine oxidoreductase (XO), which produces free radicals that trigger inflammation, atherosclerosis, aging, ischemia, hemolysis, stroke and mortality. This study aimed to analyze the effect of H. nobilis polysaccharide as an antihypercholesterolemic on the leukocyte profile of rats. This study was designed by dividing 12 male rats of the Sprague-Dawley strain into two groups. The prevention group consisted of the K+ group, the rats were fed 1% cholesterol for 28 days and the PCh group the rats were fed 1% cholesterol and H. nobilis polysaccharides at a dose of 400 mg/kg BW simultaneously for 28 days. The treatment group consisted of the Ch group, the rats were fed 1% cholesterol for the first 28 days and continued with the standard diet for the next 28 days and the ChP group, the rats were fed 1% cholesterol for the first 28 days and continued with H. nobilis polysaccharide administration of 400 mg/kg BW for the next 28 days. Hematological parameters analyzed were leukocytes, lymphocytes, neutrophils, monocytes, eosinophils, and basophils. Data analysis using One Way ANOVA and Duncant Analysis. The results showed that the number of leukocytes in the preventive control (K+) was 8883±2413 cells/mm3, PCh was 7883±728 cells/mm3, Ch was 5900±1829 cells/mm3, and ChP was 3950±975 cells/mm3. The number of lymphocytes in the K+ group was 6603±892 cells/mm3, PCh was 5070±2479 cells/mm3, Ch was 4130±1776 cells/mm3, and ChP was 2435±982 cells/mm3. The number of neutrophils in the K+ group was 2601±2258 cells/mm3, PCh was 2161±994 cells/mm3, Ch was 1593±1776 cells/mm3, and ChP was 1448±927 cells/mm3. The number of eosinophils in the K+ group was 0±0 cells/mm3, PCh was 105±451 cells/mm3, Ch was 59±146 cells/mm3, and ChP was 26±79 cells/mm3. The number of basophils in the K+, PCh, Ch, and ChP groups was 0±0 cells/mm3. The administration of sea cucumber polysaccharides H. nobilis significantly affected the number of leukocytes, in the group given H. nobilis polysaccharides leukocyte count was lower than the group not given H. nobilis polysaccharides. The sea cucumber polysaccharide H. nobilis is more effective for treatment than prevention. The administration of sea cucumber polysaccharide H. nobilis significantly affected the number of types of leukocytes.
