Title: Analisis Stabilitas Hasil dan Adaptabilitas Galur-Galur Harapan Padi Sawah Dihaploid Toleran Cekaman Salinitas

Abstract: Padi sebagai salah satu bahan pangan utama masyarakat Indonesia memerlukan peningkatan produksi seiring dengan pertambahan jumlah penduduk. Namun, tantangan peningkatan salinitas tanah akibat perubahan iklim memerlukan varietas padi yang memiliki toleransi terhadap cekaman tersebut. Program pemuliaan padi terus dilakukan sebagai upaya untuk menghasilkan varietas padi yang memiliki produktivitas tinggi dan toleran terhadap cekaman salinitas. Penelitian ini menggunakan materi genetik berupa galur-galur harapan padi sawah dihaploid hasil kultur antera. Cakupan penelitian ini meliputi uji multilokasi untuk mendapatkan informasi stabilitas hasil dan keunggulan galur-galur harapan, terutama pada karakter hasil/produktivitas dan komponen hasil serta uji toleransi terhadap cekaman salinitas pada galur-galur harapan tersebut. Luaran yang diharapkan ialah diperoleh galur-galur harapan yang memiliki potensi hasil dan produktivitas yang tinggi, stabil hasilnya, karakter komponen hasil yang baik, dan memiliki keunggulan toleran terhadap cekaman salinitas. Pengamatan pada uji multilokasi di 13 lokasi/lingkungan secara umum menunjukkan galur-galur harapan dihaploid (DH) memiliki karakter komponen hasil dan hasil/produktivitas yang tergolong baik. Sebanyak tujuh galur harapan DH memiliki rata-rata produktivitas yang lebih tinggi dibandingkan kedua varietas pembanding yang digunakan (Inpari 13 dan Inpari 30). Galur-galur harapan DH menunjukkan perbedaan peringkat hasil/produktivitas pada lokasi/lingkungan yang berbeda dan beberapa galur memiliki produktivitas yang nyata lebih tinggi pada beberapa lokasi dibandingkan semua varietas pembanding. Metode analisis stabilitas Kang, Francis-Kannenberg, Finlay-Wilkinson, Additive Main Effect and Multiplicative Interaction (AMMI), dan GGE biplot mengidentifikasi beberapa galur DH yang stabil dan berproduktivitas tinggi serta galur-galur yang beradaptasi spesifik, tetapi tidak ada galur yang teridentifikasi stabil menurut metode Eberhart-Russell. Galur HS1-5-1-1, HS17-21-1-2, HS4-13-1-4, HS4-8-1-2, HS4-13-1-3, HS4-17-1-1, dan HS15-13-1-1 merupakan tujuh galur terbaik berdasarkan keunggulannya pada potensi hasil dan produktivitas, stabilitas hasil, dan karakter komponen hasil. Penapisan terhadap cekaman salinitas dilakukan pada fase bibit menggunakan sistem hidroponik, tiga varietas pembanding toleran (Pokkali, Inpari 34 Salin Agritan, dan Inpari 35 Salin Agritan), dan satu varietas pembanding peka (Inpara 4). Pengamatan toleransi terhadap cekaman salinitas dilakukan secara kualitatif menggunakan skoring kerusakan bibit berdasarkan Standard Evaluation System (SES) for Rice dari IRRI dan berhasil mengidentifikasi 11 galur DH yang termasuk toleran dan 3 galur DH yang termasuk moderat toleran serta varietas Inpari 13 dan Inpari 30 yang termasuk moderat toleran. Pengamatan kuantitatif dilakukan pada karakter pertumbuhan bibit yang meliputi karakter tinggi bibit, panjang akar, jumlah daun, jumlah anakan, bobot basah tajuk, bobot basah akar, bobot kering tajuk, bobot kering akar, bobot basah biomassa, dan bobot kering biomassa. Karakter yang berkaitan dengan tajuk lebih dapat menggambarkan tingkat toleransi genotipe padi terhadap salinitas dibandingkan dengan karakter yang berkaitan dengan akar. Analisis lebih lanjut untuk mengidentifikasi toleransi galur-galur harapan terhadap cekaman salinitas dilakukan menggunakan analisis clustergram berdasarkan karakter seleksi yang diperoleh melalui skoring kerusakan bibit, analisis ragam, nilai duga heritabilitas, dan analisis korelasi. Penapisan toleransi menggunakan clustergram lebih ketat atau lebih selektif dibandingkan dengan hanya menggunakan skoring kerusakan bibit, sehingga diperoleh 10 galur DH tergolong toleran serta empat galur DH, Inpari 13, dan Inpari 30 tergolong peka. Sebanyak enam galur teridentifikasi memiliki potensi hasil dan produktivitas yang tinggi, karakter komponen hasil yang baik, stabil hasil/produktivitasnya, beradaptasi luas dan tergolong toleran terhadap cekaman salinitas pada fase bibit. Enam galur tersebut ialah HS1-5-1-1, HS4-13-1-4, HS4-8-1-2, HS4-13-1-3, HS4-17-1-1, dan HS15-13-1-1. Satu galur dihaploid lainnya, yaitu galur HS17-21-1-2 juga termasuk unggul berdasarkan keragaan agronomi dan stabilitas hasilnya, walaupun tergolong moderat toleran terhadap cekaman salinitas.
Rice is a staple food of most Indonesian people. The increase of rice production in Indonesia is needed due to its population growth. However, increasing soil salinity caused by climate change has become another challenge that must be solved by producing tolerant rice varieties. Rice breeding programs had been carried out to produce high yielding and salinity-tolerant rice varieties. The genetic materials used in this research were promising doubled-haploid lowland rice lines derived from anther culture. The scope of this research were multilocation trials to obtain information on agronomic characters and yield stability and adaptability of the promising lines and screening to salinity stress to obtain information on the lines’ tolerance. The expected outcomes are promising doubled-haploid lines having high productivity and yield potential, stable in yield, good yield component characters, and tolerant to salinity stress. The evaluation on multilocation trial in 13 locations/environments showed that promising doubled-haploid lines (DH lines) generally had good yield component characters. Seven DH lines showed higher average productivity than two check varieties (Inpari 13 and Inpari 30). There were changes in yield rank among DH lines across the environments and several of these lines have higher productivity than the check varieties in several environments. Stability analysis methods of Kang, Francis-Kannenberg, Finlay-Wilkinson, Additive Main Effect and Multiplicative Interaction (AMMI), and GGE biplot identified several high-yielding DH lines that were stable and identified several lines that had specific adaptation but none of the genotypes evaluated in this research were identified as stable genotypes according to Eberhart-Russell method. HS1-5-1-1, HS17-21-1-2, HS4-13-1-4, HS4-8-1-2, HS4-13-1-3, HS4-17-1-1, and HS15-13-1-1 were the seven best lines based on their superiority on yield potential, productivity, yield stability, and yield component characters. Screening to salinity stress in this research was conducted at the seedling stage using hydroponic system, three tolerant check varieties (Pokkali, Inpari 34 Salin Agritan and Inpari 35 Salin Agritan), and one sensitive check variety (Inpara 4) Salinity tolerance was assessed based on visual injury according to the Standard Evaluation System (SES) for Rice by the International Rice Research Institute (IRRI). This evaluation identified 11 promising doubled-haploid lines (DH lines) that were tolerant and three lines were moderately tolerant. Inpari 13 and Inpari 30 were identified as moderately tolerant. Quantitative evaluation was conducted based on shoot height, root length, leaf number, tiller number, shoot and root fresh weight, shoot and root dry weight, total fresh and dry weight biomass. The characters related to shoot could be better descriptors than root-related characters. Clustergram analysis based on visual injury and several selected quantitative characters indicated that 10 DH lines were categorized as moderately tolerant genotypes and four DH lines, Inpari 13, and Inpari 30 as sensitive genotypes. This analysis was more selective than that based on visual injury alone. There were six lines that had high yield potential dan productivity, good yield component characters, good yield stability, wide adaptation, and tolerance to salinity stress at the seedling stage. These lines were HS1-5-1-1, HS4-13-1-4, HS4-8-1-2, HS4-13-1-3, HS4-17-1-1, dan HS15-13-1-1. There was one DH line, i.e HS17-21-1-2, which was also superior based on its agronomic characters and yield stability but moderately salt tolerant line.
