Abstract: | Penggunaan antibiotik pada ayam petelur merupakan salah satu alternatif
untuk mengurangi kejadian penyakit; itu juga digunakan dalam aplikasi produksi
seperti kemampuan menghasilkan telur, pertumbuhan otot, dan lain-lain. Namun
penggunaan antimikroba yang tidak mengikuti aturan yang digunakan dapat
menyebabkan terjadinya resistansi mikroba. Penelitian ini bertujuan untuk
mendeteksi resistansi antibiotik terhadap gen penyandi resistansi pada isolat
bakteri Pseudomonas aeruginosa yang diisolasi dari ayam petelur di Kabupaten
Cianjur, Jawa Barat. Enam puluh enam sampel dikumpulkan dari peternakan
ayam di Kabupaten Cianjur, Jawa Barat. Sampel diidentifikasi berdasarkan
fenotipe dan genotipe. Sampel positif diuji antibiotik menggunakan metode difusi
cakram Kirby-Bauer dan gen yang mengkode resistansi dideteksi dengan PCR.
Hasil penelitian menunjukkan bahwa 8 sampel bakteri Pseudomonas aeruginosa
teridentifikasi memiliki resistansi terhadap antibiotik beta-laktam (ampisilin
75%), tetrasiklin (50% tetrasiklin, 75% oksitetrasiklin dan 50% doksisiklin),
fluoroquinolones (enrofloksasin 12,5%). ), fenikol (klorampenikol 12,5%) dan
aminoglikosida (gentamisin 0%). Deteksi gen penyandi resistansi secara berurutan
menunjukkan blaTEM (100%), blaCTXM (100%), tetA (83,3%) dan qnrS
(71,4%). Pada penelitian ini, isolat Pseudomonas aeruginosa yang diperoleh
mengalami berbagai tingkat resistansi terhadap antibiotik dan gen penyandi
resistansi. Perlu dilakukan pengawasan dan pemantauan secara berkala untuk
memastikan penggunaan antibiotik secara bijak dan penelitian lebih lanjut untuk
mendeteksi sampel dari lingkungan, tempat minum, pakan dan karyawan di
lokasi, sehingga memberikan informasi dan kajian ilmiah untuk mengatur regulasi
penggunaan antibiotik pada ternak. Using of antibiotics in laying hens is an alternative to reduce disease incidence; it is also used in production applications such as the ability to produce eggs, muscle growth, and others. However, the use of antimicrobials that are not following the rules used can lead to microbial resistance. This study aims to detect antibiotic resistance against genes encoding resistance in isolates of Pseudomonas aeruginosa bacteria isolated from laying hens in Cianjur Regency, West Java. Sixty-six samples were collected from chicken farms in Cianjur Regency, West Java. Samples were identified by phenotype and genotype. Positive samples were tested for antibiotics using the Kirby-Bauer disc diffusion method and the genes encoding resistance were detected by PCR. The results showed that 8 samples of Pseudomonas aeruginosa bacteria were identified as having resistance to betalactam antibiotics (ampicillin 75%), tetracyclines (50% tetracycline, 75% oxytetracycline and 50% doxycycline), fluoroquinolones (enrofloxacin 12.5%), phenicol (chloramphenicol 12.5%) and aminoglycosides (gentamicin 0%). Detection of resistance coding genes sequentially showed blaTEM (100%), blaCTXM (100%), tetA (83.3%) and qnrS (71.4%). In this study, the isolates of Pseudomonas aeruginosa experienced various resistance levels to antibiotics and genes encoding resistance. It is necessary to carry out regular supervision and monitoring to ensure the wise use of antibiotics and further research to detect samples from the environment, drinking places, feed and employees at the location so that it provides information and scientific studies to regulate the regulation of the use of antibiotics in livestock. |
