Abstract: | Ikan nila merupakan komoditas utama budidaya perikanan yang
dibudidayakan secara intensif dan memiliki peluang terserang penyakit salah satu nya
yang disebabkan oleh bakteri Aeromonas hydrophila. Bakteri A. hydrophila dapat
membentuk biofilm yang dapat membuat bakteri lebih resisten terhadap berbagai
kondisi lingkungan. Antibiofilm yang dihasilkan oleh bakteri Actinomycetes
mampu menunjukkan aktivitas penghambatan dan perusakan struktur biofilm dari
bakteri A. hydrophila. Penelitian ini bertujuan untuk menguji ekstrak dan supernatan
dari isolat Actinomycetes sebagai antibiofilm yang diberikan melalui pakan untuk
pencegahan infeksi bakteri A. hydrophila terhadap ikan nila (Oreochromis niloticus).
Ikan nila yang digunakan memiliki bobot awal 25,70±1,25 g dan dipelihara selama
30 hari dengan perlakuan kemudian diuji tantang serta dipelihara kembali selama
14 hari. Perlakuan yang diberikan yaitu kontrol negatif (K-), kontrol positif (K+),
ekstrak isolat Actinomycetes (EK), dan supernatan isolat Actinomycetes (SP).
Ekstrak dan supernatan dari isolat Actinomycetes sebagai antibiofilm yang di coating pada pakan mampu mencegah infeksi A. hydrophila pada ikan nila dengan
menunjukkan gambaran darah yang lebih baik dan menekan kematian dari ikan nila
setelah diinfeksi dengan A. hydrophila serta perlakuan supernatan isolat
Actinomycetes menunjukkan laju pertumbuhan harian yang lebih baik. Tilapia is the main commodity of aquaculture that is cultivated intensively and has the opportunity to be attacked by diseases, one of which is caused by the bacterium Aeromonas hydrophila. A. hydrophila can form biofilms which is make this bacteria become more resisrant to various environmental condition. Antibiofilm produced by Actinomycetes able to inhibit and destruct the biofilm structure formed by A. hydrophila. This study aimed to examine the extract and supernatant of Actinomycetes isolates as antibiofilm administered through feed for the prevention of A. hydrophila bacterial infection to tilapia (Oreochromis niloticus). The tilapia used had an initial weight of 25.70±1.25 g and was reared for 30 days with treatments then challenged and reared for 14 days. The treatments include negative control (K-), positive control (K+), Actinomycetes isolate extract (EK), and Actinomycetes isolate supernatant (SP). Extract and supernatant from Actinomycetes isolate as antibiofilm coated in feed performed capability to prevent A. hydrophila infection to tilapia by showing a better properties of tilapia from haematology and suppressing mortality assay after infection with A. hydrophila and treatment of supernatant from Actinomycetes isolate showed a better daily growth rate. |
