Title: Isolasi dan Identifikasi Staphylococcus aureus pada Vagina Sapi Simmental yang Mengalami Kawin Berulang

Abstract: Staphylococcus aureus merupakan salah satu bakteri penyebab infeksi sekunder pada sapi yang mengalami kawin berulang (repeat breeding, RB). Kawin berulang dapat menyebabkan penurunan angka kebuntingan dan kelahiran pada sapi. Tujuan dari penelitian ini adalah mengisolasi dan mengidentifikasi Staphylococcus aureus dari vagina sapi Simmental yang mengalami RB dari BPTU-HPT Padang Mengatas. Sebanyak lima spesimen bilasan vagina dikoleksi dari lima ekor sapi Simmental yang mengalami RB. Metode yang digunakan untuk identifikasi bakteri pada penelitian ini, yaitu pewarnaan Gram dan uji biokimia. Spesimen dikultur pada media blood agar (BA) dengan teknik gores T, lalu dilakukan pewarnaan Gram dan dilanjutkan dengan uji biokimia. Hasil penelitian menunjukkan bahwa tidak ditemukan bakteri Staphylococcus aureus dari kelima spesimen bilas vagina sapi yang diidentifikasi. Adapun bakteri yang teridentifikasi, yaitu bakteri genus Micrococcus dan Bacillus. Kata kunci: kawin berulang, sapi Simmental, Staphylococcus aureus
Staphylococcus aureus is a bacteria’s causes secondary infection toward the repeated breeding (RB) of cows. repeated breeding could affect the decline of pregnancy rate for the cows. This study aimed to isolate and identify the Staphylococcus aureus in Simmental’s vagina through the RB in BPTU-HPT Padang Mengatas. There were five specimens of vaginal douching collected from five Simmental cows had been through repeated breeding. There were two methods used in this research to identify the bacteria, those were Gram Staining and Biochemical test. The specimens were cultivated within the media of Blood Agar (BA) by using T-Streak technique. The next step was Gram staining, and continued by biochemical testing. The result showed that there were no Staphylococcus aureus bacteria found from those identified specimens. On the other side, the bacteria identified from those specimens were genus Micrococcus and Bacillus. Keywords: repeated breeding, Simmental cows, Staphylococcus aureus
