Abstract: Sengon (Falcataria moluccana Miq.) merupakan komoditas unggulan hutan rakyat yang berpotensi dikembangkan menjadi jenis unggulan industri perkayuan. Serangan boktor (Xystrocera festiva) menjadi ancaman besar yang dapat menurunkan produksi hingga 100%. Pengendalian molekuler yang didukung perkembangan teknologi dapat digunakan untuk mengetahui gen-gen resisten yang mendukung program pemuliaan sengon. Penelitian ini bertujuan menyusun sekuens genom referensi sengon secara de novo dan mengidentifikasi keberadaan gen Alpha-amylase inhibitor (AI) dan Trypsin inhibitor (TI) pada sekuens genom yang disusun. Penyusunan genom dilakukan dengan sequencing DNA, pengecekan kualitas reads data, genom assembly, dan anotasi genom. Ray assembler memiliki cakupan genom yang terbaik walaupun sebgaian besar gen BUSCOnya tidak lengkap diterjemahkan. Terdapat dua sekuens yang mengandung gen Alpha-amylase Inhibitor (AI) dan 19 sekuens mengandung gen Trypsin Inhibitor (TI).
Sengon (Falcataria moluccana Miq.) is a leading commodity for community forests and potential to be developed into a superior type on timber industry. The presence of boktor (Xystrocera festiva) is major threat to this commodity that can reduce production up to 100%. Molecular control supported by technological developments can be used to determine resistance genes to support breeding program. This study aims to compile de novo reference genome of sengon and identify the presence of Alpha-amylase inhibitor (AI) and Trypsin inhibitor (TI) genes in the compiled genome sequence. The genome assembly methods by DNA sequencing, quality control of reads data, genome assembly, and genome annotation. Ray assembler has the best genome coverage although the completeness of the BUSCOs gene is widely classified as missing gene. There were two sequences containing the Alpha-amylase Inhibitor (AI) gene and 19 sequences containing the Trypsin Inhibitor (TI) gene.
