Abstract: Stres merupakan keadaan ketika terjadi ketidaksesuaian antara reaksi tubuh terhadap lingkungan yang mempengaruhi keadaan fisiologi normal tubuh. Penelitian ini bertujuan mengetahui rasio neutrofil terhadap limfosit (rasio N/L) sebagai indeks stres pada tikus (Rattus norvegicus) yang diberi IgG kolostrum anti AI H5N1 terenkapsulasi. Penelitian ini menggunakan 54 ekor tikus putih jantan (Rattus norvegicus) galur Sprague dawley (SD) dengan bobot badan 200-250 gr. Tikus di bagi menjadi tiga kelompok perlakuan yaitu, kontrol negatif atau K(-) yang diberikan PBS, kontrol positif atau K(+) yang diinfeksi dengan virus AI H5N1, dan kelompok perlakuan (KP) yang diberikan mikrokapsul IgG anti AI H5N1 dan diinfeksi virus AI H5N1. Sampel darah dikoleksi secara intrakardial untuk menentukan rasio N/L pada 0, 36, dan 72 jam setelah pemberian perlakuan. Hasil menunjukkan kelompok K(+) di pengamatan yang memiliki nilai N/L sebesar 0,38 pada 36 jam setelah infeksi virus AI H5N1, sedangkan kelompok K(-) yang diberikan PBS dan KP yang diberikan mikrokapsul nilai rasio N/L lebih rendah yaitu 0,04 dan 0,12. Kelompok KP yang diinfeksi virus pada jam ke 48 pada saat pengamatan jam ke 72 mengalami kenaikan rasio N/L yaitu menjadi 0,32 sedangkan kelompok lainnya hanya 0,09 dan 0,08. Diduga infeksi virus ini menyebabkan peningkatan jumlah neutrofil dan penurunan limfosit sehingga rasio N/L meningkat, namun nilai rasio N/L masih dalam pada kisaran fisiologis normal. Berdasarkan hasil pengamatan dapat disimpulkan bahwa pemberian mikrokapsul IgG anti AI H5N1 pada tikus (Rattus norvegicus) maupun infeksi virus AI H5N1 tidak menyebabkan stres yang diindikasikan dari nilai rasio N/L berada pada kisaran fisiologis normal.
Stress is condition of a mismatch between the body's reactions to its surrounding environment which affects the body's normal physiological state. This research aims determine the neutrophils to lymphocytes ratio (N/L ratio) as an index of stress in rats (Rattus norvegicus) that were given encapsulated IgG colostrum of anti-AI H5N1. This research used 54 male white rats (Rattus norvegicus) of Sprague dawley (SD) strain with a body weight of 20 250 gr. The rats were assigned into three treatment groups; the negative control or K(-) was given PBS, the positive control or K(+) infected with AI H5N1 virus, and the treatment group (KP) was given IgG microcapsules of anti-AI H5N1 and infected with AI H5N1 virus. Blood samples was to collect intra-cardially to determine the N/L ratio at 0, 36, and 72 hours after treatment. The results showed that the K(+) group in the observation had an N/L value of 0,38 at 36 hours after infection with the AI H5N1 virus, while the K(-) group was given PBS and KP was given microcapsules had lower N/L ratio values, namely 0,04 and 0,12. The KP group that was infected with the virus at 48 hours at 72 hours of observation experience d an increase in the N/L ratio to 0,32 while the other groups were only 0,09 and 0,08. It is suspected that this viral infection causes an increase in the number of neutrophils and a decrease in lymphocytes so that the N/L ratio increases, but the value of the N/L ratio is still with in the normal physiological range. Based on the observations, it can be concluded that the administration of anti-AI H5N1 IgG microcapsules to rat (Rattus norvegicus) and infection with the AI H5N1 virus did not cause stress, which was indicated by the N/L ratio being in the normal physiological range.
