Abstract: Penelitian ini bertujuan mempelajari pengaruh beberapa kombinasi zat pengatur tumbuh pada beberapa media untuk mendapatkan kalus embriogenik bawang putih kultivar Lumbu Kuning secara in vitro. Percobaan menggunakan Rancangan Acak Lengkap satu faktor yaitu komposisi media. Terdapat 4 taraf komposisi media yaitu media A (0,1 ppm 2,4-D + 0,5 ppm Kinetin), media B (0,3 ppm 2,4-D), media C (1 ppm 2,4-D + 2 ppm Kinetin + 2 ppm IAA) dan media D (0,3 ppm 2,4-D + 0,5 ppm Kinetin) yang dikelompokkan dalam 10 kelompok sebagai ulangan sehingga terdapat 40 satuan percobaan. Pada subkultur 2 dilakukan pemindahan kalus ke media dengan mengurangi konsentrasi ZPT setengah dari awalnya di setiap media. Pada subkultur ke 3 semua kalus dipindah ke media dengan BAP 2 ppm + NAA 0,1 ppm. Hasil penelitian menunjukkan bahwa seluruh media perlakuan hampir bersamaan meginduksi kalus, dan dapat menginduksi kalus 100%. Media A, pada subkultur 3 menghasilkan ukuran kalus lebih besar dibandingkan tiga media lainnya. Kalus remah paling banyak muncul pada media B dan media C pada subkultur ketiga dengan persentase 100%. Kalus beregenerasi membentuk akar (7,5%), embrio somatik fase globular, heart, scutellum dan koleoptil, serta tunas (2,5%).
This research aimed to study the effect of media compositions to obtain garlic embryogenic callus of Lumbu Kuning cultivar. The experiment is designed using a completely randomized design with a single factor, that was the composition of the media. There are 4 levels of media composition, media A (0,1 ppm 2,4-D + 0,5 ppm Kinetin), media B (0,3 ppm 2,4-D), media C (1 ppm 2,4-D + 2 ppm Kinetin + 2 ppm IAA), and media D (0,3 ppm 2,4-D + 0,5 ppm Kinetin) were grouped into 10 groups as replications so that there were 40 experimental units.In subculture 2, callus was transferred to the media by reducing the ZPT concentration by half from the beginning in each media. in the 3rd subculture, all callus was transferred to the media with BAP 2 ppm + NAA 0,1 ppm. The results showed that all treatment media induced callus almost simultaneously, and could induce callus 100%. Media A, in subculture 3 produced a largest callus size than the other three media. Crumb callus mostly appeared on media B and media C in the third subculture with a percentage of 100%. Callus regenerated to form roots (7,5%), somatic embryos in globular phase, heart, scutellum and coleoptile, also shoots (2,5%).
