Abstract: Tingginya kadar kolesterol di dalam darah menjadi salah satu faktor resiko penyebab penyakit jantung dan stroke. Belum ada informasi ilmiah yang mendukung khasiat penurunan kolesterol menggunakan kombinasi ekstrak daun jati belanda dengan daun salam secara in vitro. Penelitian ini bertujuan menguji efektivitas interaksi kombinasi ekstrak berbasis daun jati belanda sebagai penghambat aktivitas HMG-KoA reduktase. Penghambatan enzim diukur berdasarkan pengurangan NADPH menggunakan spektrofotometer λ 333 nm. Hasil menunjukkan bahwa kombinasi fraksi polifenol pada perbandingan 1:1 (50 μg/mL) memiliki daya penghambatan tertinggi yaitu 66.63%, sama baiknya dengan kombinasi ekstrak kasar 1:1 (60.24%) dan lovastatin komersial 5 μg/mL (65.52%). Penambahan daun jati belanda yang lebih banyak pada perbandingan 2:1 dan 3:1 pada kombinasi ekstrak kasar maupun fraksi polifenol melemahkan penghambatan enzim dengan nilai persen inhibisi yang lebih rendah yaitu pada kisaran 20.85%-52.22%.
High levels of cholesterol in the blood become one of the risk factors causing heart disease and stroke. There was no scientific information that supports the efficacy of cholesterol reduction using the extract combination of Guazuma ulmifolia leaves with bay leaves in vitro. The research objective of this study was to examine the effectifity of the interaction by Guazuma ulmifolia leaves based combined extractas an inhibitor of HMG CoA reductase activity. Enzyme inhibition was measured by determination of decreasing NADPH with a spectrophotometer λ 333 nm. The results showed that the combination of polyphenol fractions at the ratio of 1:1 had the highest inhibitory effect 66.63%, as well as to the combination of 1:1 crude extract (60.24%) and commercial lovastatin 5 μg/mL (65.52%). Addition of more Guazuma ulmifolia leaves at the ratio of 2:1 and 3:1 to the combination of crude extracts and polyphenol fractions reduced the inhibition of enzymes with a lower percent inhibition value on the range of 20.85%-52.22%.
