dc.description.abstractKitolod (Hippobroma longiflora (L.) G.Don) merupakan tumbuhan liar yang banyak dimanfaatkan oleh masyarakat sebagai obat tradisional. Bila pemanfaatan tanaman ini semakin berkembang, dikhawatirkan akan terjadi kekurangan ketersediaan sumber benih. Penelitian ini bertujuan mendapatkan teknik dan kondisi kultur terbaik dalam perbanyakan tanaman obat kitolod secara in vitro agar tersedia bibit yang dapat digunakan oleh masyarakat secara luas. Metode penelitian menggunakan rancangan acak lengkap (RAL) dengan dua faktor perlakuan dan 10 ulangan untuk semua percobaan kecuali pengakaran tunas adventif. Daun dan petiol kitolod digunakan sebagai bahan eksplan. Eksplan daun pada perlakuan BAP 1 mg/L + NAA 0,1 mg/L memberikan jumlah tunas adventif per eksplan paling tinggi yaitu sampai 49 tunas, namun perlakuan BAP 2 mg/L + NAA 0,1 mg/L lebih efektif dalam inisiasi dan perbanyakan tunas kitolod. Tunas-tunas pada BAP 2 mg/L + NAA 0,1 mg/L juga mampu menghasilkan tunas tertinggi (2,62±0,69 cm) pada media pembesaran dan memberikan 75% keberhasilan dalam tahap aklimatisasi. Media pengakaran terbaik adalah media MS yang ditambah NAA 0,5-1,0 mg/L.id
dc.description.abstractKitolod (Hippobroma longiflora (L.) G. Don) is a wild plant that widely used by the community as a traditional medicine. If this plant is utilized more intensively, there may be a shortage of the plant due to the lack of the seed sources. This study aimed to obtain the best techniques and culture conditions for in vitro propagation of kitolod to provide a large number of planting materials for the community. The experiments were arranged using a completely randomized design (CRD) with two treatment factors and 10 replications for all experiments except shoot rooting. Leaves and petioles were used as explant sources. Leaf explants in 1 mg/L BAP + 0,1 mg/L NAA treatment produced the highest number of adventitious shoots per explant, up to 49 shoots, but 2 mg/L BAP + 0,1 mg/L NAA was more effective for shoot initiation and multiplication. The latter medium was also able to produce the tallest shoots (2,62±0,69 cm) and presented 75% of successful rate over the acclimatization period. The best rooting medium was MS medium added with 0,5-1,0 mg/L NAA.id
