The research was undergone to study the effect of antiviral Quercetin, temperature, and repetitive subculturing on the geminivirus titer in tobacco tissue culture in order to have virus free plant. Tobacco plantlet was grown in Murashige-Skoog media with specific treatment, i.e. addition of Quercetin 10 mg. 1-1 and 20 mg. 1-1, heat therapy at 35 °C, 40 °C, and 50 °C, and two time subculturing. Measurement of virus titer was conducted serologically using Indirect ELISA method. In general, the growth of tobacco plantlets was not effected by Quercetin, nor temperature except for heat therapy at 50 °C. Addition of Quercetin to the growing media and its combination with subculturing reduced the titer of geminivirus. The same result was observed with heat therapy at 35 °C and 40 °C, and its combination with subculturing. Tissue culture plants should be tested through aclimatitation to asure the plants has been freed from systemic infection of the virus.
