Genetic transformation of indica rice by crIA(b) and cryIB gene using Agrobacterium tumefaciens to obtain yellow stem borer resistance (Scirpophaga incertulas Walker.)
Abstract
Serangkaian penelitian telah dilakukan dengan tujuan untuk mendapatkan padi indica yang tahan hama penggerek batang melalui introduksi gen cryLA(b) dan cryLB ke dalam kalus embriogenik dengan menggunakan teknik Agrobacterium tumelaciens. A series of experiment was done to obtain indica rice resistance to stem borer by cryIA(b) and cryIB genes were introduced into rice embryogenic calli using Agrobacterium tumefaciens technique. The research consisted of four steps: 1) optimation of in-vitro culture regeneration of indica rice as transformation target via Agrobacterium tumefaciens, 2) study of several vectors and transformation methods via Agrobacterium tumefaciens of indica rice, 3) construction of recombinant plasmids containing cryIA(b) and cryIB genes which suitable for Agrebacterium transformation, and 4) transformation of recombinant plasmids carrying cryIA(b) and cryIB genes to indica rice using Agrobacterium tumefaciens to obtain yellow stem borer resistance (Scirpophaga incertulas Walker.)
