      Abstract
      The aims of this study were to describe the genetic variation of D-loop of mtDNA which is usefuldatabase for conducting conservation programe. The whole blood samples were collected (8 samples for D-loop analysis) from four locations which were Aceh Besar, Pidie, North Aceh regencies and Banda Aceh city. Out group whole blood samples were collected from two samples from Bali cattles (Bali Island),Madura cattle (Madura Island), and Pesisir cattle (West Sumatera) respectively and one sample from PO cattle (West Java). Amplification of D-loop sequences of mtDNA with BIDLF and BIDLR primary had PCR product 980 bp length and sequences length which could be analyzed 479 bp. The Data were analyzed using Squint 1.02 and MEGA4.0 programme. The result of D-loop mtDNA analysis as showed that there were 27 sites of variance in Aceh cattle. This marker can be used to differentiate and subdivide of Indonesia local cattle, and indicated that Aceh cattle had the same cluster as Pesisir cattle where PO cattle comes into their group, has closer genetic distance to Bos indicus, while Bali and Madura cattle had the same cluster
       
      Penelitian yang bertujuan untuk mengkarakterisasi keragaman daerah D-loop mtDNA yang berguna sebagai database dalam pelaksanaan program konservasi sapi Aceh telah dilakukan. Pengumpulan sampel darah (8 sampel untuk analisis D-loop) dilakukan di empat lokasi yaitu Kota Banda Aceh, Kabupaten Aceh Besar, Pidie dan Aceh Utara. Sampel pembanding telah diambil 2 sampel masing-masing sapi Bali (Pulau Bali), Madura (Pulau Madura), Pesisir (Sumatera Barat) dan satu sampel sapi PO (Jawa Barat). Primer yang digunakan untuk mengamplifikasi sekuen daerah D-loop mtDNA adalah BIDLF dan BIDLR dengan panjang produk 980 bp dan panjang sekuen yang dapat dianalisis 479 bp. Data sekuen D-loop disejajarkan berganda dengan sekuen acuan Bos indicus dari GenBank menggunakan program Squint 1.02 dan dianalisis dengan program MEGA4. Hasil analisis D-loop mtDNA dengan sekuen acuan Bos indicus, ada 27 situs beragam pada sapi Aceh. Penanda ini dapat digunakan untuk membedakan dan pengelompokan sapi lokal Indonesia di mana sapi Aceh satu klaster dengan sapi Pesisir dan PO, mempunyai jarak genetik yang lebih dekat dengan sapi Bos indicus, sedangkan sapi Bali dan Madura membentuk klaster sendiri
