      Identifikasi trikoma kelenjar untuk produksi Artemisinin pada artemisia annua l.menggunakan Pendekatan molekular

      Abstract
      Produksi artemisinin yang merupakan zat bioaktif antimalaria pada tanaman Artemisia annua (Asteraceae) disekresikan oleh trikoma kelenjar yang telah mencapai kematangan fisiologi. Penelitian ini bertujuan untuk melihat pada tahap pertumbuhan daun yang dapat menghasilkan trikoma kelenjar yang sudah mencapai kematangan fisiologi (berkembang sempurna) dalam jumlah besar serta melihat ekspresi dari gen CYP71AV, gen yang bertanggung jawab dalam biosintesis artemisinin. Pengamatan trikoma kelenjar pada beberapa tahapan pertumbuhan daun (kuncup, setengah membuka, lamina berkembang sempurna, dan daun tua sebelum gugur) menunjukkan bahwa baik pada aksesi ungu (genjah) maupun aksesi hijau (dalam) produksi artemisinin tertinggi terdapat pada stadia perkembangan daun setengah membuka dan ditandai dengan banyaknya trikoma kelenjar yang belum pecah. Ekspresi gen CYP71AV tertinggi pada aksesi hijau terdapat mulai daun kuncup dan maksimal pada daun setengah membuka, kemudian menurun pada daun membuka dan tidak terekspresi pada daun luruh. Sedangkan pada aksesi ungu ekspresi tertinggi hanya terdapat pada daun kuncup dan sedikit pada daun membuka. Berdasarkan hasil yang diperoleh maka dianjurkan untuk memanen tanaman sebelum antesis pada stadia daun setengah membuka sampai membuka
       
      Artemisia annua (Asteraceae) has a glandular trichome which secretes artemisinin, antimalarial substances. This research aimed to study the relation between glandular trichomes maturity with leaf developmentand expression of CYP71AV gene that responsible to biosynthesis of artemisinin during leaf development. Observation on development of glandular trichomes was done before anthesis (maximum vegetative growth) and at fourth stage of leaf development. Glandular trichomes in leaves in fourth stage development (leaf tip, half open leaf, maturity leaf, fall leaf) were studied. It has been observed that number of glandular trichomes were increased from half leaf open stage until leaf maturity stage in green accession, but in purple accession was increased until fall leaf stage. Artemisinin content analysis showed that half open leaf on both accession had higher content of artemisinin compare than other stage of leaves. Expression of CYP71AV gene was increased from leaf tip until half leaf open stage and decreased during leaf maturity and no expression on the fall leaf stage in green accesion. But,in purple accesion expression of CYP71AV gene only on half leaf open stage. Based on this results, it was suggested to harvest leaves of A. annua at half leaf open stage before anthesis.
