Aplikasi Marka RAPD dalam Seleksi Galur Kedelai Toleran Naungan
Abstract
The increase of soybean consumption in Indonesia not been met by domestic soybean production due to low productivity and limited planting area leading to a large amount of import. Expansion of planting area beyond the traditional soybean planting area as intercrop with estate crops is an alternative to reduce import. The main constraint in planting soybean as intercrop with estate crops is low light intensity in addition to drought, low soil pH and Aluminium toxicity. Breeding for soybean varieties with improved adaptation to low light intensity under intercropping condition is currently in progress at the Center for Crop Improvement Studies, Department of Agronomy and Horticulture, Bogor Agricultural University. Selection of RAPD markers was conducted with 85 random primers using Bulk Segregate Analysis. Phenotype of recombinant in bred lines (RILs) for the BSA was conducted under artificial shading. From polimorphic bands, several markers, OPM 19, OPH07, OPE 19, OPH2 and OPM8, SBOI-22, G10, I09, P01 were linked to tolerant lines. These marker were then validated using tolerant lines selected from intercropping with rubber trees. The two markers were consistently linked to tolerant lines, namely OPM 19 nnd P01, and were used in the marker assited selection of 300 soybean lines from BP2TP and IPB
