      Effectivity of Immunoglobulin Y Antilipase as Pancreatic Lipase Inhibitor for Prevention of Obesity

      Abstract
      Obesity has been a major risk for many degenerative diseases with an increasing prevalence.Pancreatic lipase is responsible for 50-70% of total dietary fat absorbtion and has been studied widely for new anti-obesity invention. Hen immunized with porcine pancreatic lipase will produce yolk containing IgY antilipase. IgY antilipase will bind the active site of pancreatic lipase in gastrointestinal tract and inhibit the activity of pancreatic lipase in the gastrointestinal tract. This research was conducted to evaluate the effectivity of IgY antilipase as anti-obesity. IgY antilipase was purified with sodium sulphate precipitation. The protein was analysed by SDS-PAGE and was identified by Immunoblotting. The ability of IgY antilipase to inhibit pancreatic lipase was tested by colorimetric microplate assay at five grades of concentration. Yolk containing IgY antilipase was then tested in rabbit to evaluate its ability to inhibit fat absorbtion in vivo. The result showed that IgY contained two proteins with molecular weights of 61.2 kDa and 26.9 kDa. IgY was confirmed because it positively reacted with rabbit-anti-chicken HRP. The average IgY concentration was 30.1 ± 6.3 mg/mL yolk. IgY antilipase and orlistat did not differ in inhibition capacity at various concentrations (p<0.05). The inhibition capacity of IgY obtained from unimmunized hen was significantly lower as compared to those of IgY antilipase and orlistat (p<0.05). Yolk containing IgY antilipase also had the ability to inhibit dietary fat absorbtion in vivo, but its inhibition capacity was lower than that of orlistat (p<0.05). It is concluded that IgY antilipase could be used to inhibit fat absorbtion to prevent obesity.
