Fractionation and identification of cytotoxic components from binahong (Anredera cordifolia) leaves based on Artemia salina Leach analysis have been carried out. Air-dried binahong leaves were macerated in ethanol as solvent. The ethanol crude extract was further partitioned with n-hexane, dichloromethane, and n-butanol. Toxicity test with A. salina showed that the n-butanol extract was the most toxic with 50% lethal concentration (LC50) of 494μg/mL. Fractionation of the n-butanol extract by using column chromatography and gradient elution with n-hexane-chloroform-methanol resulted 5 fractions. Fraction 4 was the most toxic with LC50= 23.4μg/mL, and gave single spot in thin layer chromatography with chloroform-methanol (7:3) as eluent. Phytochemistry test results and analysis of Fourier transform infrared spectrum showed that the active components of the fraction had carboxyl, aromatic ring, and ester groups. Gas chromatography-mass spectrometry analysis obtained bis-2-ethylhexyl-1,2-benzenedicarboxylate as the main component (79.29%), along with oleic acid (5.30%) and palmitic acid (4.78%).
