Anodonta woodiana is one of freshwater mussel which have been widely cultivated for its ability to produce pearls. A. woodiana has the ability to produce nacre and prismatic crystal that produce pearls. This ability is closely related to genetic information in this species. The aim of this study was to examine the characteristic of cytochrome oxidase subunit I (COI) gene in freshwater mussel A. woodiana from West Java Province. Genomic DNA was extracted from tissue of A. woodiana from Bogor and Depok. Extracted DNA continued by amplification and sequencing of COI gene. COI gene was aligned with some COI genes from other species, ingroup (familiy Unionidae) and outgroup obtained from GenBank. Amplified of COI gene were successfully yielded in 608 bp. This study was not able to characterize COI gene of A. woodiana well because there are inhibitors. Nucleotid along 608 bp can not be used as a barcode. Phylogenetic analysis of A. woodiana showed that A. woodiana is separated from other family Unionidae.
