Antidementia activity of the Syzygium cumini, S. aromaticum, and S. polyanthum leaves, as well as S. aromaticum buds extracts were determined based on inhibition of acetylcholinesterase (AChE) activity mechanism. The aim of this study was to determine the most potent AChE inhibitor. Each samples were extracted by using sonication methods with n-hexane, ethyl acetate, and methanol as solvents. The inhibition activity was measured with modified Ellman’s method by using microplate reader spectrophotometer at 412 nm. The methanol extract of S. aromaticum leaves, S. polyanthum leaves, S. aromaticum buds, and the ethyl acetate extract of S. polyanthum leaves were potential as AChE inhibitors. The 50% inhibition concentration (IC50) values were 42.13, 47.15, 45.22, and 45.02 μg/mL, respectively, while the IC50 value of physostigmine was 0.01 μg/mL. Thin layer chromatography fingerprint pattern of the extracts eluted with dichloromethane:ethyl acetate (1/2:1/2) as the best eluent and silica gel GF254 as stationary phase show 9, 7, 2, and 5 separated peaks, respectively.
