      Abstract
      Begomoviruses has been reported to cause serious diseases in a number of economically crops in the world. Infection of Begomovirus in Indonesia was reported since 1999 and caused significant yield losses especially in chillipepper. The objectives of this research were to isolate, characterize and clone coat protein gene from Begomovirus infecting chillipepper into pGEM-T Easy vector and to use the clone as DNA Probe in dot blot hybridization. Coat protein gene of Begomovirus isolate originated from Ngluwar-Magelang was successfully isolated, characterized and cloned to pGEM-T Easy. Based on multiple alignment analysis using clustal W it is evidenced that recombinant DNA of Begomovirus isolate Ngluwar has high homology (> 90%, e value 0,0) with other Begomoviruses published earlier in GenBank. Dot blot hybridization method using recombinant DNA clone of Begomovirus isolate Ngluwar as DNA probe was able to detect Begomovirus from various sources (infected plants, PCR products from total DNA, cloned DNA) with sensitivity up to 0,0001 μg/μl. The use of DNA probe should be further evaluated for its use as detection tool for Begomovirus.
       
      Infeksi Begomovirus (Famili Geminiviridae) dilaporkan telah menyebabkan kehilangan hasil yang tinggi pada beberapa jenis tanaman. Di Indonesia serangan Begomovirus pada pertanaman cabai pertama kali dilaporkan pada tahun 1999 di daerah Jawa Barat oleh Hidayat et al. (2006). Selanjutnya Sulandari et al. (2006) melaporkan bahwa kejadian penyakit yang tinggi pada pertanaman cabai yang disebabkan oleh Begomovirus berlangsung sejak awal tahun 2000, telah menyebabkan kehilangan hasil yang tinggi di beberapa daerah yaitu: Daerah Istimewa Yogyakarta, Jawa Tengah dan Jawa Barat, dengan kehilangan hasil pada cabai besar mencapai 100% dan pada cabai kecil 50-70%. Hingga saat ini Begomovirus masih menjadi patogen penting yang menyebabkan kehilangan hasil yang tinggi pada pertanaman cabai di Indonesia. Begomovirus penyebab penyakit pada tanaman cabai di beberapa daerah di Indonesia tersebut selanjutnya disebut Pepper Yellow Leaf Curl Indonesia Virus (PYLCIV). Karakteristik Begomovirus yang tinggi turut menentukan sulitnya mengendalikan serangan penyakit pada berbagai tanaman inangnya yaitu keragaman genetiknya yang tinggi. Implikasi dari keragaman tersebut bermanfaat dalam pengembangan metode deteksi Begomovirus yang tepat dan cepat mendeteksi keberadaan Begomovirus.
