Asam gelugur fruits (Garcinia atroviridis) and kunci pepet rhizomes (Kaempferia rotunda) are examples of traditional medicinal plants that have potential as antiobesity. Mechanism inhibition of these herbals in reducing body weight has not been known yet. The objective of this research is to evaluate the mechanism inhibition of these herbals as antiobesity on pancreatic lipase enzyme. Water extract of asam gelugur fruits contains saponins and the ethanol extract contains alkaloids. Water extract of kunci pepet rhizomes contains alkaloids, flavonoids, saponins, and tannins, while the ethanol extract contains alkaloids, flavonoids, saponins, and triterpenoids. Pancreatic lipase enzyme kinetics pattern formed after addition of the ethanol extract of asam gelugur fruits caused an increase in KM in by to 0.0626 ppm and vmax values decreased by 9.0853 ppm-/-min. Kinetics of lipase pancreatic enzyme after addition of the water extract of kunci pepet rhizomes increased the KM value by 0.0694 ppm and vmax values decreased by 3.1760 ppm-/-min. The kinetics of inhibition test showed that the mechanism inhibition tend to be competitive mechanism inhibition based on the increasing value of KM. Buah asam gelugur (Garcinia atroviridis) dan rimpang kunci pepet (Kaempferia rotunda) merupakan contoh tanaman obat tradisional yang berpotensi sebagai antiobesitas. Mekanisme hambatan kedua tanaman ini dalam menurunkan bobot badan belum diketahui dengan pasti. Penelitian ini bertujuan mengevaluasi mekanisme hambatan kedua tanaman sebagai antiobesitas terhadap enzim lipase pankreas. Ekstrak air buah asam gelugur mengandung saponin dan ekstrak etanolnya mengandung alkaloid. Ekstrak air rimpang kunci pepet mengandung alkaloid, flavonoid, saponin, dan tanin, sedangkan ekstrak etanolnya mengandung alkaloid, flavonoid, saponin, dan triterpenoid. Pola kinetika enzim lipase pankreas yang terbentuk setelah penambahan ekstrak etanol buah asam gelugur menyebabkan peningkatan KM sebesar 0.0626 ppm dan penurunan nilai vmaks sebesar 9.0853 ppm/menit. Kinetika enzim lipase pankreas setelah penambahan ekstrak air rimpang kunci pepet mengalami peningkatan KM sebesar 0.0694 ppm dan penurunan nilai vmaks sebesar 3.1760 ppm/menit.
