Pengaruh konsentrasi colchicine terhadap pertumbuhan bibit tumbuhan pulai (Alstonia scholaris R. Br.)
Abstract
Pulai is one of useful plant in Indonesia. This plant has efficacy as a herb. Use of traditional herbs currently tends to increase. To anticipate the increasing use of traditional herbs and exploitation, it is necessary to improve the quality and production of medicinal plant cultivation. It also required to increase the benefits of pulai so it is possible to keep its sustainability. To improve the quality and value of pulai benefits is by providing chemical colchicine which has the ability to form poliploid plants. This research aims to examine the effect of colchicine concentration on the growth of pulai seedling on nurseries media. The research was conducted in Tissue Culture Laboratory, Research Center for Science Bogor Agriculture University from May to August 2010. This research was compiled using statistical methods of RAL One Factorial. There are four treatments using colchicine with different concentrations and control. Colchicine concentration used was 0.5 mg/l, 1.0 mg/l, 1.5 mg/l, and 2.0 mg/l. Each treatment had 10 replications for a total of 50 units. Colchicine was given until three days consecutively, then controlled the plant for two weeks as a break. The treatment was repeated six times. Observation was done in one week during the break, for all experimental units include its parameters such as the growth of height of shoot, diameter, and number of leaves. Plant cultivation includes the provision of green hyponex fertilizer, hormones of the shoot, bactericide, fungicide, amoxycilin, and weeding. The results of this research indicate that the highest average height of shoot and diameter is found at a concentration of 1.0 mg/l, while the average of highest number of leaves is found on control plants. Overall, the result of statistical test on three parameters is not significantly different, but it can not be denied from a possibility of poliploid. The conclusion of this research is the concentration of colchicine in this research does not provide a significantly different effect on the growth of seedlings pulai, but it can not be denied that there was a tendency of poliploid on seedlings given treatment.
