      Correlation of in vitro germination and staining method in pollen viability testing

      Abstract
      Pollen viability of four families were investigated based on in vitro germination and staining method and then data were correlated. The aim was to study if the two methods gave similar estimation of pollen viabilities. Pollen germination medium consisted of Brewbaker & Kwack solution (BK), BK without Sucrose, Sucrose 10%, and modified pollen germination medium formulated by Schreiber and Dresselhaus (2003) (PGM). Staining method consists of aniline blue, acetocarmine, IKI, and TTC. Pollen morphology varies among the four families but very similar between species within a family. Euphorbiaceae pollen (d= 81,2µm/J. pandurifolia) and Poaceae (d= 48,9µm/S. bicolor) was round with one pore (uniporate). Solanaceae have an oval shape with three colpates pores (tricolpate). In the other hand, Myrtaceae pollen varies either triangle with tricolpate or square with four colpates. The best germination medium for almost all species except in Myrtaceae was PGM, and the best stain was acetocarmine. However, corellation between PGM and acetocarmin was not significantly correlated, but PGM was correlated with aniline blue (r=0.624) meaning that aniline blue could be used to estimate pollen viability as good as in vitro germination using PGM..
