      Abstract
      Conventional cancer treatments can induce immunosupression and other side effects that lead to progressive tumors growth and reccurence. It has become the reason of choosing traditional medicines as antitumor therapy. The aim of this research is to observe the antiproliferation activities of temulawak ethanol extract on MCA-B1 and MCM-B2 tumor cell lines in vitro. Cells were cultivated in tissue culture plate 24 well with vinblastin (positive control) and different concentrations of extract in 5 replicates. The concentrations were 0 ppm (negative control), 15 ppm (P1), 30 ppm (P2), 45 ppm (P3), 60 ppm (P4), and 75 ppm (P5). Cells were incubated at 370C, 5% CO2. After confluence had been attained on control wells (day third), cells were harvested and total cells were counted using a haemocytometer Neubauer. The results showed that temulawak ethanol extract had antiproliferation activities on MCA-B1 and MCM-B2 tumor cell lines. The best result was given by 75 ppm dose, with antiproliferation activity reached for 70.0% on MCA-B1 cells and 75.4% on MCM-B2 cells. Based on all results, we concluded that temulawak ethanol extract have a possible therapeutic potential against cancer and suggested that this plant extract could be developed as an anticancer substance.
       
      Pengobatan kanker secara konvesional dapat menimbulkan imunosupresi dan efek samping lainnya yang memberikan peluang bagi pertumbuhan tumor yang progresif atau timbulnya rekurens. Oleh karena itu, pengobatan tradisional menjadi salah satu alternatif pilihan. Penelitian ini bertujuan untuk mengetahui adanya aktivitas antiproliferasi ekstrak etanol temulawak terhadap pertumbuhan sel lestari tumor MCA-B1 dan MCM-B2 secara in vitro. Penelitian dilakukan dengan menanam sel lestari tumor MCA-B1 dan MCM-B2 pada tissue culture plate 24 well dengan vinblastin (kontrol positif) dan konsentrasi ekstrak yang berbeda sebanyak 5 kali ulangan. Konsentrasi ekstrak yang digunakan adalah 0 ppm (kontrol negatif), 15 ppm (P1), 30 ppm (P2), 45 ppm (P3), 60 ppm (P4), dan 75 ppm (P5). Sel diinkubasikan pada suhu 370C, 5% CO2. Pemanenan dilakukan setelah confluence, yaitu pada hari ketiga dan penghitungan jumlah sel dilakukan dengan hemositometer Neubauer. Hasil penelitian menunjukkan adanya aktivitas antiproliferasi ekstrak etanol temulawak pada sel lestari tumor MCA-B1 dan MCM-B2. Dosis ekstrak yang memberikan hasil paling baik adalah 75 ppm dengan aktivitas antiproliferasi sebesar 70,0% pada sel lestari tumor MCA-B1 dan 75,4% pada sel lestari tumor MCM-B2. Hasil tersebut menunjukkan potensi temulawak sebagai tanaman obat antitumor dan kami menyarankan agar tanaman ini dapat dikembangkan sebagai salah satu obat antitumor.
