      KARAKTERISASI VIRUS PENYAKIT MULUT DAN KUKU ISOLAT DARI PROVINSI JAWA BARAT TAHUN 2022-2023

      Abstract
      Penyakit mulut dan kuku (PMK) adalah infeksi virus akut dan sangat menular yang menyerang berbagai hewan berkuku genap/belah. Pada tahun 2022, PMK re-emerging di Indonesia, dengan wabah yang dilaporkan di beberapa provinsi, termasuk Jawa Barat. Kemunculan kembali ini telah menimbulkan kekhawatiran tentang pengelolaan dan pengendalian penyakit, terutama mengingat karakteristik virus yang kompleks. Virus PMK menunjukkan karakteristik yang berbeda berdasarkan serotipe. Studi ini bertujuan untuk mengidentifikasi gejala klinis PMK pada beberapa jenis hewan yang rentan, mendeteksi, mengisolasi, dan mengkarakterisasi virus PMK penyebab wabah 2022-2023 di Jawa Barat. Studi ini mengumpulkan 31 sampel dari Kabupaten Sumedang dan Sukabumi di Provinsi Jawa Barat, yang terkena wabah PK antara tahun 2022 dan 2023. Pemeriksaan klinis dilakukan pada hewan yang diduga terinfeksi virus PMK; 31 sampel tersebut terdiri dari 20 sapi, 9 kerbau, dan 2 kambing. Pemeriksaan laboratorium yang digunakan adalah deteksi molekuler dengan RT-qPCR, isolasi dan serial passage, identifikasi dengan RT-PCR, dan karakterisasi molekuler dengan sekuensing. Gejala klinis yang diamati meliputi demam (52%), salivasi dan keluarnya cairan hidung berlebihan (45%), lepuh di mulut (42%), pincang (23%), dan lesi kaki (16%). Hasil RT-qPCR dari 30 dari 31 sampel (97%) menunjukkan positif virus PMK. Total 30 sampel positif PMK dengan RT-qPCR, 8 sampel diantaranya diidentifikasi sebagai serotipe O dengan ukuran amplikon RT-PCR 1135 bp. Virus PMK berhasil dikultur dalam sel BHK-21 dan menunjukan efek sitopatik (kematian atau kerusakan sel) dalam waktu 24–72 jam. Serial passage dan analisis sekuensing protein VP1 pada passage 1 (P1), passage 5 (P5), dan passage 10 (P10) menunjukkan mutasi titik substitusi. Titik mutasi P1 di protein terletak pada basa 618; P2 pada basa 618, 624, dan 667; P3 pada basa 618, 667, dan 790. Gejala klinis penyakit mulut dan kuku (PMK) yang diamati pada sapi, kerbau, dan kambing di Provinsi Jawa Barat konsisten dengan yang dilaporkan sebelumnya. Virus PMK terdeteksi pada 97% hewan yang menunjukkan gejala klinis dalam penelitian ini.
       
      Foot and mouth disease (FMD) is an acute and highly contagious viral infection that affects various cloven-hoofed animals. In 2022, FMD re-emerged in Indonesia, with outbreaks reported in several provinces, including West Java. This resurgence has raised concerns about the management and control of the disease, especially given the virus's complex characteristics. FMD virus exhibits distinct characteristics based on serotypes. This study aims to identify clinical symptoms of FMD in different species, to detect, to isolate and to characterize the FMD virus causing the 2022-2023 outbreak in West Java. This study collected 31 samples from Sumedang and Sukabumi districts in West Java Province, which were affected by FMD outbreaks between 2022 and 2023. Clinical examinations were conducted on animals suspected of being infected with the FMD virus; the 31 samples comprised 20 cattle, 9 buffaloes, and 2 goats. The study method used was molecular detection with RT-qPCR, isolation and serial passage, identification with RT-PCR, and molecular characterization with sequencing. The observed clinical symptoms included fever (52%), excessive salivation and nasal discharge (45%), vesicles in the mouth (42%), lameness (23%), and leg lesions (16%). The RT-qPCR result of 30 out of 31 samples (97%) tested positive for FMD virus. Of the 30 samples that were positive for FMD, 8 were identified as serotype O, with an RT-PCR amplicon size of 1135 bp. FMD virus was successfully cultured in BHK-21 cells, producing a cytopathic effect (CPE) within 24–72 hours. Serial passage and sequencing analysis of the VP1 protein at passage 1 (P1), passage 5 (P5), and passage 10 (P10) revealed point substitution mutations. The point mutations in P1 in the VP1 protein are located at base 618; P2 at bases 618, 624, and 667; and P3 at bases 618, 667, and 790. The clinical symptoms of FMD observed in cattle, buffalo, and goats in West Java Province are consistent with those previously reported. FMD virus was detected in 97% of animals showing clinical signs in this study.
