      Validasi Metode Penentuan Besi dan Zink dalam Suplemen Herbal Peningkat Imunitas dengan Spektrofotometer Serapan Atom

      Abstract
      Keberadaan mineral besi (Fe) dan zink (Zn) dalam suplemen herbal peningkat imunitas perlu diawasi untuk memastikan keamanan dan efektivitas penggunaannya. Penentuan kadar Fe dan Zn dilakukan dengan metode spektrofotometer serapan atom (SSA), yang melibatkan proses preparasi destruksi basah untuk Fe menggunakan HCl, dan destruksi kering untuk Zn. Validasi metode mencakup aspek-aspek seperti spesifisitas, uji kesesuaian sistem, linearitas, limit deteksi, limit kuantisasi, presisi, akurasi, ketahanan, dan ketangguhan yang sesuai dengan standar nasional dan internasional. Hasil pengujian menunjukkan linearitas dengan nilai r² mendekati 1, serta akurasi dengan persentase perolehan kembali sebesar 98,37–100,98% untuk Fe dan 98,53–102,62% untuk Zn. Kadar Fe dan Zn dalam sampel berturut-turut adalah 15,04 mg/tablet dan 9,71 mg/tablet. Analisis statistik uji ketahanan dan ketangguhan menunjukkan bahwa F-hitung < F-tabel dan t-hitung < t-tabel, yang mengindikasikan bahwa metode ini stabil dan valid untuk analisis mineral dalam suplemen herbal.
       
      The presence of iron (Fe) and zinc (Zn) in immune-enhancing herbal supplements was monitored to ensure their safety and efficacy. The determination of Fe and Zn levels was carried out using atomic absorption spectrometer (AAS), involving wet digestion with hydrochloric acid for Fe and dry ashing for Zn. Method validation covered parameters including specificity, system suitability, linearity, limit of detection, limit of quantitation, precision, accuracy, robustness, and ruggedness, in accordance with national and international standards. The results showed good linearity with r² approaching 1, and acceptable accuracy with recovery percentages of 98.37–100.98% for Fe and 98.53–102.62% for Zn. The Fe and Zn contents in the samples were 15.04 mg/tablet and 9.71 mg/tablet, respectively. Statistical evaluation of robustness and ruggedness showed that F-calculated < F-table and t-calculated < t-table, indicating the method was stable, and valid for mineral analysis in herbal supplements.
