      Analisis Fisikokimia dan Aktivitas Antibakteri Madu Hutan Timor Terhadap Methicillin-Resistant Staphylococcus aureus

      Abstract
      Kemunculan bakteri yang resistan antibiotik telah menjadi perhatian dunia. Madu memiliki potensi sebagai agen alternatif dalam mengatasi resistansi antibiotik. Penelitian ini bertujuan untuk menganalisis sifat fisikokimia, kandungan hidrogen peroksida (H2O2), pinocembrin, kaempferol, serta aktivitas antibakteri madu hutan Timor. Sebanyak tujuh sampel madu hutan Timor dianalisis untuk mengetahui kadar air, aktivitas air, pH, H2O2, pinocembrin, kaempferol, dan aktivitas antibakterinya. Hasil penelitian menunjukkan bahwa rata-rata kadar air, aktivitas air, dan pH madu hutan Timor masing-masing adalah 17,52%, 0,6, dan 4,18. Kadar air, aktivitas air, dan pH diukur menggunakan refraktometer, aw-meter, dan pH meter secara berturut-turut. Kandungan hidrogen peroksida dianalisis menggunakan peroxide strip test. Kandungan pinocembrin dan kaempferol dianalisis menggunakan High Performance Liquid Chromatography-Diode Array Detection (HPLC-DAD). Aktivitas antibakteri terhadap Methicillin-Resistant Staphylococcus aureus (MRSA) dievaluasi menggunakan metode difusi cakram Kirby-Bauer, dengan vankomisin sebagai kontrol positif. Kandungan H2O2, pinocembrin, dan kaempferol berturut-turut adalah 109,1 µmol/L, 2,2 mg/100 g, dan 1,7 mg/100 g. Daya hambat madu hutan Timor dan vankomisin terhadap MRSA tidak berbeda nyata dengan rata-rata diameter zona hambat 13,93±0,23 mm untuk madu hutan Timor dan 14,33±0,05 mm untuk vankomisin. Madu hutan Timor memiliki karakteristik fisikokimia dan senyawa bioaktif yang berkontribusi terhadap aktivitas antibakterinya. Temuan ini menunjukkan potensi madu hutan Timor sebagai agen alami dalam upaya mengatasi infeksi yang disebabkan oleh bakteri resistan terhadap antibiotik.
       
      The emergence of antibiotic-resistant bacteria has become a global concern. Honey has the potential to serve as an alternative agent in combating antibiotic resistance. This study aimed to determine the physicochemical properties, hydrogen peroxide (H2O2), pinocembrin, kaempferol and antimicrobial activity of Timor forest honey. Seven samples of Timor forest honey were analyzed for water content, water activity, pH, H2O2, pinocembrin, kaempferol and antibacterial activity. Moisture content, water activity, and pH were measured using a refractometer, an aw-meter, and a pH meter, respectively. Hydrogen peroxide levels were assessed using peroxide strip test. Pinocembrin and kaempferol contents were analyzed using High Performance Liquid Chromatography with Diode Array Detection (HPLC-DAD). The results showed that the average water content, water activity and pH of Timor forest honey were 17,52%, 0,6, 4,18, respectively. The levels of H2O2, pinocembrin, and kaempferol were 109,1 µmol/L, 2,2 mg/100 g, 1,7 mg/100 g, respectively. Antibacterial activity against Methicillin-Resistant Staphylococcus aureus (MRSA) was evaluated using Kirby-Bauer disc diffusion method, with vancomycin as a positive control. The inhibition zone of Timor forest honey and vancomycin were not significantly different with mean diameters of 13,93±0,23 mm and 14,33±0,05 mm, respectively. Timor forest honey possesses physicochemical properties and bioactive compounds that contribute to its antibacterial activity. These findings highlight its potential as a natural alternative for combating antibiotic-resistant bacteria.
