      Karakteristik dan Kinematika Pergerakan Spermatozoa Domba dalam Medium Fertilisasi dan Kajian Kemampuan Fertilisasinya Secara in Vitro

      Abstract
      Kualitas spermatozoa merupakan faktor penting dalam mendukung keberhasilan fertilisasi in vitro. Penelitian ini bertujuan mengevaluasi efektivitas sericin terhadap kualitas dan kinematika spermatozoa dalam medium fertilisasi dan sebagai pengganti BSA pada teknologi produksi embrio in vitro. Persiapan spermatozoa dilakukan dengan dan tanpa swim up. Setiap kelompok diinkubasi dalam media fertilisasi dengan BSA, sericin 0,05%, 0,1%, dan 0,5% serta dievaluasi segera setelah ditambahkan media fertilisasi (menit ke-0), menit ke-15, 30 dan 60. Setiap akhir masa inkubasi, kualitas dan kinematika dievaluasi sebanyak empat ulangan. Hasil penelitian menunjukkan bahwa motilitas progresif kelompok sericin 0,5% lebih tinggi dibanding kelompok lain (P<0,05). Motilitas menurun signifikan setelah terpapar media fertilisasi, kecuali perlakuan sericin 0,5% yang menurun pada menit ke-30. Integritas membran spermatozoa menunjukkan perbedaan antar perlakuan, kecuali menit ke-30 dan 60 kelompok swim up perlakuan sericin 0,5% tidak berbeda dengan BSA dan sericin 0,1%. Nilai VCL menunjukkan tidak terdapat perbedaan kecuali kelompok swim up menit ke-0 dan 30 nilai sericin 0,5% lebih tinggi (P<0,05). VAP, VSL, LIN dan STR tidak terdapat perbedaan, namun LIN dan STR kelompok swim up pada menit ke-30 nilai sericin 0,05% lebih rendah (P<0,05). Nilai ALH kelompok swim up menit ke-30 dan menit ke-60 tanpa swim up sericin 0,5% lebih tinggi dibanding perlakuan lainnya (P<0,05). Simpulan penelitian ini, penambahan sericin 0,5% berpotensi menggantikan BSA serta dapat mempertahankan kualitas dan menunjukkan tanda awal hiperaktivasi motilitas spermatozoa. Berdasarkan studi literatur disimpulkan bahwa penambahan sericin pada media maturasi, fertilisasi, kultur serta ditambahkan saat kriopreservasi spermatozoa dapat meningkatkan kualitas fertilisasi dan perkembangan embrio pada berbagai spesies hewan.
       
      Spermatozoa quality is an important factor in the success of in-vitro fertilization. This study aimed to evaluate the effectiveness of sericin on spermatozoa in fertilization medium and as a substitute for BSA in embryo production technology. Spermatozoa were prepared with and without swim-up. Each group was incubated in fertilization medium with BSA, sericin 0.05%, 0.1%, and 0.5%, evaluated immediately after adding fertilization medium (0th minute), 15, 30 and 60. At the end of the incubation period, quality and kinematics were evaluated in four replicates. The results showed that the progressive motility of sericin 0.5% was higher (P<0.05). Motility decreased significantly after exposure to fertilization media, except sericin 0.5%, which decreased at 30 minute. Membrane integrity showed differences between treatments, except for 30 and 60 minutes of swim-up group, sericin 0.5% treatment did not differ from BSA and sericin 0.1%. The VCL showed no difference except for the 0 and 30 minute swim-up groups, sericin 0.5% value was higher (P<0.05). VAP, VSL, LIN, and STR had no difference, but LIN and STR of the swim-up group at 30 minute sericin 0.05% had lower value (P<0.05). The ALH of the 30 minute swim-up and 60 minute without swim-up sericin 0.5% were higher (P<0.05). The conclusion is sericin 0.5% has potential to replace BSA, maintain quality, and show early signs of hyperactivation. Based on literature studies, the addition of sericin to maturation media, fertilization, culture, and spermatozoa cryopreservation can improve the quality of fertilization and embryo development in various animal species.
