      Aplikasi Bakteri Endofit secara In Vitro untuk Meningkatkan Ketahanan Bawang Merah terhadap Penyakit Layu Fusarium dan Bercak Ungu

      Abstract
      Penyakit utama yang menyebabkan permasalahan pada budi daya bawang merah disebabkan oleh Fusarium oxysporum dan Alternaria porri. Alternatif lain untuk pengendalian penyakit layu Fusarium dan bercak ungu dapat dilakukan dengan teknik pengendalian biologis yang memanfaatkan bakteri endofit. Penelitian ini bertujuan mengetahui strain bakteri endofit yang dapat meningkatkan ketahanan bawang merah terhadap serangan penyakit bercak ungu yang disebabkan oleh A. porri dan penyakit layu Fusarium yang disebabkan oleh F. oxysporum. Rancangan percobaan yang digunakan dalam penelitian ini adalah Rancangan Acak Lengkap (RAL) faktor tunggal. Perlakuan yang diberikan adalah inokulasi kandidat bakteri endofit strain 35, 36, 37, 39, G053, 2, 16 dan 19.2. Uji ketahanan dilakukan pada media seleksi yaitu media kultur bawang merah yang diberi filtrat F. oxysporum 30% dan A. porri 20%. Pengamatan dilakukan terhadap pertumbuhan tunas, daun, dan akar, gejala serangan pathogen dan jumlah umbi. Hasil penelitian menunjukkan bakteri strain 35 mampu menekan keparahan penyakit sebanyak 56% pada filtrat A. porri dan 69% pada filtrat F. oxysporum, bakteri strain 39 sebanyak 57% pada filtrat A. porri dan 66% pada filtrat F. oxysporum, dan bakteri strain G053 sebanyak 69% pada filtrat A. porri dan 36% pada filtrat F. oxysporum.
       
      The main diseases that cause problems in shallot cultivation are caused by Fusarium oxysporum and Alternaria porri. Another alternative to control Fusarium wilt and purple spot disease can be done with biological control techniques that utilise endophytic bacteria. This study aims to determine the strain of endophytic bacteria that can increase shallot resistance to purple spot disease caused by A. porri and Fusarium wilt disease caused by F. oxysporum. The experimental design used in this study was a single-factor completely randomised design (CRD). The treatments given were inoculation of endophytic bacterial candidates strains 35, 36, 37, 39, G053, 2, 16 and 19.2. The resistance test was carried out on selection media, namely shallot culture media containing 30% F. oxysporum filtrate and 20% A. porri. Observations were made on the growth of shoots, leaves, and roots, symptoms of pathogen attack and the number of bulbs. The results showed that strain 35 bacteria were able to suppress disease severity by 56% in A. porri filtrate and 69% in F. oxysporum filtrate, strain 39 bacteria by 57% in A. porri filtrate and 66% in F. oxysporum filtrate, and strain G053 bacteria by 69% in A. porri filtrate and 36% in F. oxysporum filtrate.
