      Pengaruh 2,4-D dan Kinetin dalam Induksi Kalus Eksplan Ruas Vanili (Vanilla Planifolia Andrews.) var. Vania 1 secara In Vitro

      Abstract
      Organogenesis melalui kalus diharapkan meningkatkan keragaman genetik vanili melalui induksi keragaman somaklonal. Penelitian ini bertujuan mempelajari pengaruh pemberian 2,4-D dan kinetin serta menentukan konsentrasi yang tepat dalam menginduksi kalus dari ruas vanili var. Vania 1 secara in vitro.Penelitian dilakukan dengan Rancangan Kelompok Lengkap Teracak (RKLT) faktorial dengan dua faktor perlakuan. yaitu konsentrasi 2,4-D (0; 0,5; 1,0; 1,5 mg L-1 ) dan kinetin (0; 0,05; 0,1 mg L -1 ) yang diulang sebanyak 10 kali sehingga terdapat 120 satuan percobaan. Hasil penelitian menunjukan bahwa kombinasi 2,4-D dan kinetin tidak berpengaruh signifikan terhadap persentase eksplan hidup dan browning serta ukuran kalus, namun interaksi keduanya dan pengaruh tunggal 2,4- D berpengaruh signifikan terhadap persentase eksplan berkalus. Kombinasi 2,4-D 1,5 mg L -1 dan kinetin 0,05 mg L -1 menghasilkan persentase eksplan berkalus tertinggi sebesar 22,22%. Pemberian 2,4-D konsentrasi 1,5 mg L?¹ secara tunggal menghasilkan persentase kalus tertinggi sebesar 14,36%, sementara pemberian kinetin dengan konsentrasi 0,05 mg L?¹ menghasilkan waktu tercepat untuk munculnya kalus yaitu 13,94 hari. Kalus dapat beregenerasi membentuk tunas pada kombinasi 2,4-D 1 mg L-1 dan kinetin 0,05 mg L-1 . Kalus yang terbentuk memiliki tekstur kompak dengan variasi warna dari pale yellow (p.Y) hingga moderate orange yellow (M.OY).
       
      Organogenesis through callus formation is expected to enhance the genetic diversity of vanilla through the induction of somaclonal variation. This study aimed to examine the effects of 2,4-D and kinetin application and determine the optimal concentrations for inducing callus from vanilla var. Vania 1 internode in vitro. The experiment was conducted using a factorial Randomized Complete Block Design (RCBD) with two factors: 2,4-D concentrations (0, 0.5, 1.0, 1.5 mg L?¹) and kinetin concentrations (0, 0.05, 0.1 mg L?¹), replicated 10 times, resulting in 120 experimental units.The results showed that the combination of 2,4-D and kinetin did not significantly affect explant survival percentage, browning, or callus size. However, their interaction and the single effect of 2,4-D significantly influenced the percentage of callus-forming explants. The combination of 2,4-D 1.5 mg L?¹ and kinetin 0.05 mg L?¹ produced the highest percentage of callus-forming explants at 22.22%. The application of 2,4-D at a concentration of 1.5 mg L?¹ alone resulted in the highest callus percentage at 14.36%, while kinetin at a concentration of 0.05 mg L?¹ produced the shortest time for callus initiation, at 13.94 days. Callus regeneration into shoots was observed in the combination of 2,4-D 1 mg L?¹ and kinetin 0.05 mg L?¹. The resulting callus exhibited a compact texture with color variations ranging from pale yellow (p.Y) to moderate orange yellow (M.OY).
