      Regenerated Silk Fibroin dari Serat Sutra dan Karakterisasinya

      Abstract
      Dalam beberapa tahun terakhir, fibroin dari serat sutra semakin diminati sebagai biomaterial untuk rekayasa jaringan karena biokompatibilitas, biodegradabilitas, dan kekuatan tariknya yang tinggi. Untuk diaplikasikan, fibroin harus dilarutkan dalam pelarut tertentu menjadi regenerated silk fibroin (RSF). Metode pelarutan fibroin dari ulat sutra B. mori sudah banyak diteliti, namun sedikit yang dilakukan pada ulat S. ricini yang lebih mudah dikembangkan di Indonesia. Penelitian ini bertujuan mencari kondisi optimal pelarutan fibroin B. mori dan S. ricini serta karakterisasinya. Pelarut ionik diketahui dapat melarutkan fibroin tanpa merusak struktur primernya. Fibroin B. mori mudah larut dalam LiBr, reagen Ajisawa (CaCl2–H2O–EtOH), dan Ca(NO3)2 menghasilkan larutan dengan konsentrasi 7,19%, sedangkan fibroin S. ricini hanya larut dalam Ca(NO3)2 setelah 34 jam pada 90 °C dan menghasilkan larutan dengan konsentrasi 8,90%. Proses liofilisasi menghasilkan serbuk RSF putih pada B. mori dan coklat pada S. ricini. Spektrum FTIR menunjukkan sebagian besar struktur koil acak dan sebagian kecil lembaran-ß pada RSF. Hasil ini menunjukkan keberhasilan pelarutan fibroin pada B. mori dan S. ricini.
       
      In recent years, silk fibroin has gained interest as a biomaterial for tissue engineering due to its biocompatibility, biodegradability, and high tensile strength. To apply it, fibroin must be dissolved in specific solvents to form regenerated silk fibroin (RSF). While methods for dissolving fibroin from B. mori silkworms are well-established, little research has been done on S. ricini, which is easier to cultivate in Indonesia. This study aims to find optimal conditions for dissolving B. mori and S. ricini fibroin and characterizing the RSF. Fibroin can be dissolves without damaging its primary structure using ionic solvents. B. mori fibroin dissolves easily in LiBr, Ajisawa reagent, and Ca(NO3)2, producing a solution with a concentration of 7.19%. Meanwhile, S. ricini fibroin only dissolves in Ca(NO3)2 after 34 hours at 90 °C, resulting in a solution with a concentration of 8.90%. The lyophilization produced white RSF powder for B. mori and brown for S. ricini. FTIR spectra showed RSF is primarily composed of random coils and some ß-sheets structures. These results demonstrate successful fibroin dissolution for both B. mori and S. ricini.
