      Karakterisasi Aktivitas Proteolitik Dari Fraksi Ekstraseluler Isolat Bakteri Bacillus cereus SLBD Dari Kotoran Kuda

      Abstract
      Bacillus cereus SLBD (Bc-SLBD) sebelumnya diisolasi dari kotorankudadan menunjukkan potensi sebagai produsen enzim proteolitik untuk berbagai aplikasi. Namun, karakteristik dasar dari bakteri ini masih belumdipahami denganbaik. Penelitian ini bertujuan untuk menyelidiki karakteristik dasar Bc-SLBD, termasuk morfologi, perilaku pertumbuhan, dan produksi proteolitiknya. Morfologi Bc-SLBD diperiksa menggunakan pewarnaan Gramdan mikroskopelektron, yang mengungkapkan bahwa bakteri ini bersifat Gram-positif danberbentuk batang. Analisis lebih lanjut mengenai perilaku pertumbuhan dalammedia Luria-Bertani (LB) broth pada berbagai suhu menunjukkan bahwa Bc- SLBD memiliki suhu pertumbuhan optimal pada 37 °C. Aktivitas protease, yangdinilai dengan mengukur diameter zona bening pada berbagai fase pertumbuhan, menunjukkan bahwa fase stasioner adalah yang paling optimal untuk produksi protease. Hasilnya juga menunjukkan bahwa aktivitas protease Bc-SLBDoptimal pada suhu 50 °C dan pH 8,0. Penelitian ini memberikan dasar untuk produksi danaplikasi lebih lanjut dari protease SLBD
       
      Bacillus cereus SLBD (Bc-SLBD) was previously isolated fromhorsefeces and shows promise as a producer of proteolytic enzymes for variousapplications. However, the basic characteristics of this bacteriumare still not well understood. This study aims to investigate the fundamental characteristics of Bc- SLBD, including its morphology, growth behavior, and proteolytic production. The morphology of Bc-SLBD was examined using Gram staining and scanningelectron microscopy, revealing that this bacterium is Gram-positive androd- shaped. Further analysis of growth behavior in Luria-Bertani (LB) brothat different temperatures indicated that Bc-SLBD has an optimal growth temperatureof 37 °C. Protease activity, assessed by measuring the clear zone diameter at different growth phases, showed that the stationary phase is the most optimal for production. The results also demonstrate that the protease activity of Bc-SLBDisoptimal at a temperature of 50 °C and a pH of 8.0. This study provides afoundation for the further production and application of SLBD’s proteases.
