      Pengaruh Jenis Media Pertumbuhan terhadap Hasil Pengujian Angka Lempeng Total pada Lada Hitam

      Abstract
      GLORYA T.O. LUBIS. Pengaruh jenis media pertumbuhan terhadap hasil pengujian angka lempeng total pada lada hitam. Dibimbing oleh DWI YUNI HASTATI Lada hitam merupakan jenis lada yang dipanen dan dijemur ketika buah lada sudah mendekati waktu matang. Kontaminasi mikroba menjadi salah satu masalah mutu mikrobiologi lada hitam dan dapat diuji menggunakan metode angka lempeng total (ALT) yang merupakan parameter umum dalam memperlihatkan kontaminasi mikroba pada pangan. ALT dapat diuji menggunakan media Plate Count Agar (PCA), Nutrient Agar (NA) dan Tryptone Soya Agar (TSA). Tujuan penelitian adalah mengetahui pengaruh media pertumbuhan (PCA, NA, TSA), bentuk butir dan bubuk serta sumber sampel (petani, pedagang, eksportir) terhadap ALT pada lada hitam. Hasil penelitian menunjukkan bahwa media pertumbuhan tidak berbeda nyata terhadap ALT lada hitam dengan nilai signifikansi 0,095. Bentuk dan sumber sampel berbeda nyata terhadap ALT lada hitam dengan nilai signifikansi bentuk sampel adalah 0,004 dan nilai signifikansi sumber sampel adalah 0,000. Sampel berbentuk butir memiliki ALT lebih tinggi yaitu 1,2 × 106 cfu/g dan sampel dari eksportir memiliki ALT paling rendah yaitu 8,7 × 104 cfu/g. Berdasarkan data, pengujian ALT lada hitam dapat menggunakan tiga media tersebut. Kata kunci: angka lempeng total, bentuk, lada hitam, media pertumbuhan, sumber sampel
       
      Black pepper is a type of pepper that is harvested and dried when the pepper fruit is nearing maturity. Microbial contamination is one of the microbiological quality problems of black pepper and can be tested using the total plate count (TPC) method which is a common parameter in showing microbial contamination in food. TPC can be tested using Plate Count Agar (PCA), Nutrient Agar (NA) and Tryptone Soya Agar (TSA) media. The purpose of this study was to determine the effect of growth media (PCA, NA, TSA), grain and powder forms and sample sources (farmers, traders, exporters) on TPC in black pepper. The results showed that the growth media did not differ significantly to TPC in black pepper with a significance value of 0,095. The shape and source of the sample were significantly different to TPC in black pepper with a significance value of sample form of 0,004 and a significance value of sample source of 0,000. The granular sample had a higher ALT of 1,2 × 106 cfu/g and the sample from the exporter had the lowest TPC of 8,7 × 104 cfu/g. Based on the data, black pepper TPC testing can use these three media.
