      Produksi Pigmen Pyocyanin dari Pseudomonas aeruginosa dan Aktivitas Ekstraknya sebagai Antibakteri Xanthomonas oryzae pv. oryzae

      Abstract
      Pigmen pyocyanin yang dihasilkan oleh bakteri Pseudomonas aeruginosa dapat berfungsi sebagai agen antimikrob terhadap beberapa mikrob patogen. Namun, biaya produksi yang tinggi dan rendemen yang rendah menyebabkan produksi pyocyanin dalam skala besar masih sangat terbatas. Penelitian ini bertujuan menganalisis potensi beberapa jenis media alternatif dalam mendukung pertumbuhan dan produksi pyocyanin asal P. aeruginosa, dan mengevaluasi potensi pyocyanin sebagai agen biokontrol mikrob fitopatogen. Terdapat dua isolat dengan kode P1.S9 dan D2.Z13 yang dipakai dalam penelitian ini. Kedua isolat P. aeruginosa memiliki perbedaan warna kultur. Isolat P1.S9 memiliki warna biru kehijauan, sedangkan D2.Z13 hijau kekuningan. Berdasarkan hasil optimasi menggunakan media alternatif, konsentrasi pyocyanin tertinggi adalah sebesar 2,584 ± 0,053 µg/ml diperoleh dari P. aeruginosa P1.S9 yang ditumbuhkan pada media rebusan singkong (200 g/L). Meskipun konsentrasi pyocyanin yang diperoleh masih jauh dari media standar King’s A (14,415 ± 0,146 µg/ml), namun hal tersebut telah membuktikan potensi penggunaan media alternatif untuk produksi pyocyanin. Hal tersebut diperkuat dengan nilai aktivitas antibakteri pyocyanin asal dua media tersebut yang tidak jauh berbeda. Ekstrak pyocyanin isolat P1.S9 yang diekstrak dari media King’s A dan rebusan singkong memiliki aktivitas antibakteri terhadap Xanthomonas oryzae pv. oryzae dengan zona hambat berkisar antara 6,94-10,28 mm dan 7,45 mm.       Pyocyanin pigment produced by Pseudomonas aeruginosa bacteria can function as an antimicrobial agent against several pathogenic microbes. However, high production costs and low yields cause large-scale pyocyanin production to be very limited. This study aims to analyze the potential of several types of alternative media in supporting the growth and production of pyocyanin from P. aeruginosa, and to evaluate the potential of pyocyanin as a biocontrol agent for phytopathogenic microbes. There are two isolates with codes P1.S9 and D2.Z13 used in this study. Both P. aeruginosa isolates have different culture colors. Isolate P1.S9 has a greenish blue color, while D2.Z13 is yellowish green. Based on the results of optimization using alternative media, the highest pyocyanin concentration of 2.584 ± 0.053 µg/ml was obtained from P. aeruginosa P1.S9 grown on cassava decoction (200 g/L). Although the concentration of pyocyanin obtained is still far from the standard King's A medium (14.415 ± 0.146 µg/ml), it has proven the potential of using alternative media for pyocyanin production. This is reinforced by the antibacterial activity values of pyocyanin from the two media which are not much different. Pyocyanin extract of isolate P1.S9 extracted from King's A medium and cassava decoction has antibacterial activity against Xanthomonas oryzae pv. oryzae with inhibition zones ranging from 6.94-10.28 mm and 7.45 mm.
