      Abstract
      Penyimpanan dan pemeliharaan koleksi kultur mikrob adalah elemen penting dalam studi sistematika dan keanekaberagaman hayati. Mengingat keberagaman yang tinggi pada mikrob, diperlukan berbagai metode pemeliharaan dan pelestarian untuk memastikan kelangsungan hidup serta integritas morfologi, fisiologis, dan genetik kultur dari waktu ke waktu. Penelitian ini bertujuan untuk menginventarisasi, merangkum dan menjabarkan beberapa temuan hasil penelitian penelitian sebelumnya terkait beragam cara koleksi, penyimpanan dan pemeliharaan isolat mikrob disertai contoh langsung hasil penelitian. Penelitian ini dilaksanakan dengan studi pustaka pada bulan Januari-Juli 2024 dan pengaplikasian teknik isolasi mikrob pada bulan Juni hingga September 2022 di Laboratorium Bioteknologi Perairan, Departemen Teknologi Hasil Perairan, Institut Pertanian Bogor. Hasil kajian ini menyimpulkan bahwa sumber sampel koleksi mikrob bisa diperoleh dari beragam sampel seperti tanah, air, tanaman, udara, sementara teknik pemeliharaan isolat mikrob bisa dilakukan diantaranya dengan cara penyimpanan berkala, penyimpanan dalam aquades steril, penyimpanan dalam tanah steril dan penyimpanan dalam minyak mineral.
       
      Culture collection and preservation of microbial culture is an important element in systematics and biodiversity studies. The high diversity of microbes, a variety of cultivation and preservation methods are required to ensure survival and morphological, physiological, and genetic integrity of cultures over time. This research aims to inventory, summarize and describe several findings from previous research related to various methods of collecting, storing and maintaining microbial isolates along with direct examples of research results. This research was carried out by literature study during January to May 2024 and application of microbial isolation techniques from June to September 2022 at the Aquatic Biotechnology Laboratory, Department of Aquatic Products Technology. Institut Pertanian Bogor. The results of this study concluded that the source of microbial collection samples can be obtained from various samples such as soil, water, plants, airborne spores. Meanwhile, preserving techniques for maintaining microbial isolates can be performed by periodic storage, storage in sterile distilled water, storage in sterile soil and storage in mineral oil.
