      Extract of Dumb Cane (Dieffenbachia seguine (Jacq.) Schott) as a Potential Inflammatory Inducer in Wistar Rats

      Abstract
      Dieffenbachia seguine (Jacq.) Schott is a species of tropical flowering plant in the family of Araceae. In the West, this plant is known as Dumb Cane, while in Indonesia it is referred to as Sri Rejeki. It has a poisoning effect of raphides, such as needle-shaped calcium oxalate crystals, that can be causing temporary swelling. This research aimed to study the efficacy of Dieffenbachia seguine extract as an inflammatory inducer in Wistar rats as an animal model and compare it with carrageenan. This research used 24 male Wistar rats and divided them into 6 treatment groups, namely, normal control (Water for Injection/WFI), base, positive control (carrageenan 1%), 1%, 2%, and 4% D. seguine extract emulsion. Using a mercury plethysmometer, paw volume was measured just before the induction, at “0 hours” and then at 1, 2, 3, 4, and 24th hours after the intraplantar injection. Data were analyzed using one-way ANOVA with Duncan's test. The results showed that 2% and 4% extract emulsion could induce an inflammatory response in Wistar rats through intraplantar injection and were not significantly different from 1% carrageenan. The most optimal swelling occurs in the 4th hour after induction. 2% D. seguine extract showed the most efficient effect in inducing inflammatory response among the three concentration extracts.
       
      Dieffenbachia seguine (Jacq.) Schott adalah tanaman hias daerah tropis yang termasuk dalam family Araceae. Di Negara Barat, tanaman ini dikenal dengan nama Dumb Cane sedangkan di Indonesia adalah Sri Rejeki. Dieffenbachia seguine memiliki senyawa racun kristal kalsium oksalat berbentuk jarum (raphides) yang dapat menyebabkan pembengkakan sementara. Penelitian ini bertujuan untuk mengetahui efek ekstrak Dieffenbachia seguine sebagai agen induksi inflamasi pada hewan model tikus Wistar dan membandingkannya dengan karagenan. Penelitian ini menggunakan 24 ekor tikus Wistar jantan dan dibagi menjadi 6 kelompok perlakuan yaitu kontrol normal (Water for Injection/WFI), basis, kontrol positif (karagenan 1%), serta emulsi ekstrak konsentrasi 1%, 2% dan 4%. Volume kaki diukur sesaat sebelum induksi, pada “0 jam” dan kemudian pada 1, 2, 3, 4, dan 24 jam setelah injeksi intraplantar menggunakan merkuri plethysmometer. Data dianalisis menggunakan one-way ANOVA dengan uji lanjut Duncan. Hasil penelitian menunjukkan emulsi ekstrak konsentrasi 2% dan 4% dapat menginduksi respon inflamasi pada tikus Wistar melalui injeksi intraplantar dan tidak berbeda signifikan dengan karagenan 1%. Kebengkakan paling optimal terjadi pada jam ke-4 setelah induksi. Ekstrak D. seguine 2% menunjukkan efek yang paling efisien dalam menginduksi respon inflamasi di antara ketiga ekstrak konsentrasi.
