Analisis In Silico Biflavonoid dari Genus Araucaria sebagai Inhibitor Protein DENV NS5 RNA-dependent RNA polymerase (RdRp) Menggunakan Aplikasi PLANTS

Demam berdarah dengue (DBD) adalah penyakit yang ditularkan oleh nyamuk Aedes aegypti. Infeksi virus dengue ini disebabkan adanya produksi virus DENV pada protein NS5 RNA-dependent RNA polymerase. Balapiravir merupakan senyawa yang mampu menjadi inhibitor NS5 RdRp tetapi tidak dapat menurunkan viremia. Genus Araucaria mengandung senyawa biflavonoid yang telah diteliti berpotensi sebagai antivirus melalui inhibisi NS5 RdRp. Penelitian ini bertujuan memprediksi 23 senyawa turunan biflavonoid dari genus Araucaria sebagai antidengue melalui inhibisi NS5 RdRp berdasarkan parameter penambatan molekuler dengan aplikasi PLANTS, serta hubungan aktivitas dan strukturnya. Aplikasi PLANTS didasarkan pada perilaku koloni semut yang bergerak di sekitar situs pengikatan untuk menemukan konformasi ligan terbaik. Hasil penelitian menunjukkan bahwa terdapat 6 senyawa yang berpotensi sebagai inhibitor NS5 RdRp dengan skor penambatan lebih rendah dibandingkan balapiravir. Senyawa yang paling berpotensi, yaitu senyawa 4’,4’’’-di-O-metil kupresuflavon (22), dengan skor penambatan -96,0786. Dengue fever (DHF) is a disease transmitted by the Aedes aegypti mosquito. Dengue virus infection is caused by the production of DENV virus on the NS5 RNA-dependent RNA polymerase protein. Balapiravir is a compound that can be an inhibitor of NS5 RdRp but cannot reduce viremia. The Araucaria genus contains biflavonoid compounds that have been studied as potential antivirals through NS5 RdRp inhibition. This study aims to predict 23 biflavonoid-derived compounds from the Araucaria genus as antidengue through NS5 RdRp inhibition based on molecular docking parameters with the PLANTS application, as well as activity and structure relationships. The PLANTS application is based on the behavior of ant colonies that move around the binding site to find the best ligand conformation. The results showed that there are 6 compounds that have potential as NS5 RdRp inhibitors with lower docking scores than balapiravir. The most potential compound is 4',4''di-O-methyl-cupresuflavone (22), with a docking score of -96.0786.
